川芎嗪逆转K562/ADM细胞多耐药性的研究

摘　　要：目的 :研究具有钙拮抗作用的中药川芎嗪对多药耐药性的逆转作用。方法 :选用异搏定作阳性对照 ,观察川芎嗪 10 μg/m L 剂量下对多药耐药细胞株 K5 6 2 /A DM多药耐药性的逆转作用。结果 :川芎嗪在非细胞毒性剂量下能使 K5 6 2 /ADM对阿霉素和长春新碱的 IC5 0 降低 ,使细胞内阿霉素和柔红霉素的浓度升高 ,但对细胞表面的糖蛋白 P-170却没有影响。结论 :川芎嗪具有逆转白血病细胞株 K5 6 2 /ADM多药耐药性的作用。Objective: It is to study the reverse effect of ligustrazine, a kind of resisting calici herb medicine, multidrug resistance(MDR). Methods:Verapamil was selected as positive control. The effect of ligustrazine 10μg/ml on reversing MDR of K562/ADM cell was observed. Results:Ligustrazine could make MDR of cell line of K562/ADM reduce the value of IC 50 of Adriamycin and Vincristine against at non cytotoxic concentration and enhance density of Adriamycin and Daunorubicin in cell, but cause no change on expression of P 170. Conclusion:Ligustrazine can reverse MDR of leukemic cell line of K562/ADM.
