摘 要:【目的】研究日本血吸虫卵黄铁蛋白DNA免疫在两种处理小鼠诱导的免疫应答及其保护作用。【方法】免疫小鼠分两大组进行 :实验Ⅰ组小鼠有预感染 ,实验Ⅱ组小鼠没有预感染。将构建的携带日本血吸虫卵黄铁蛋白基因的重组质粒pLXSN Fer1经左后腿肌肉注射BALB/c小鼠 ,共免疫 2次 ,间隔 2周。末次免疫后 5周以血吸虫尾蚴攻击感染 ,6周后剖杀小鼠 ,计数成虫负荷及肝组织虫卵数。设空载质粒免疫组为对照组。【结果】免疫小鼠后在实验Ⅰ组主要诱生IgG1和IgG3,在实验Ⅱ组主要诱生IgG2a和IgG2b ;实验组产生较高水平的IgA和IFN γ应答。pLXSN Fer1免疫小鼠获得一定程度的减虫率 (2 6 47% ,34 0 8% )和减卵率 (4 0 0 2 % ,36 83% )。【结论】日本血吸虫卵黄铁蛋白在两种不同处理小鼠均能产生一定程度的保护作用 ,有可能成为有价值的疫苗候选分子 。To study the immune responses to Shistosoma japonicum yolk ferritin DNA vaccination in mice with two approaches of pretreatments and its protecting effects. Set two series of immunization experiments. Designed beforehand infection in group Ⅰ, but not in group Ⅱ. BALB/c mice were vaccinated with pLXSN Fer1 encoding yolk ferritin gene of Chinese Schistosoma japonicum to each quadriceps muscle of left leg. Each group was immunized twice with 2 weeks interval. Mice vaccinated with pLXSN blank vector served as control. Mice were challenged five weeks after final DNA boosting by percutaneous infection with cercariae. Six weeks after infection the mice were perfused, worm burden and eggs in liver were counted. Sera from mice were collected from tail vein at weeks 3,11,17 in group Ⅰ, and weeks 0,6,12 in group Ⅱ,respectively. In group Ⅰ, mice produced predominantly IgG1 and IgG3; whereas in group Ⅱ, IgG2a and IgG2b antibodies. Mice vaccinated with pLXSN Fer1 also produced IgA and IFN γ. Immunization with pLXSN Fer1 in the two groups could achieve significant worm reduction rate(26 47%, 34 08%) and egg reduction rate(40 02%, 36 83%).[Conclusion] DNA vaccination with pLXSN Fer1 could induce protective immunity in BALB/c mice significantly. It may serve as a moleculer of a potential vaccine and be worth of further research.
