摘  要:以普通念珠藻为原料 ,用甲醇脱脂 ,再用水作提取剂 ,控制温度 98℃ ,提取 3次 ,每次 3 8h ,合并提取液 ,减压浓缩 ,用蛋白酶法脱蛋白 1次 ,Sevag法脱蛋白 7次 ,用蒸馏水透析 2d ,醇析后冷冻干燥 ,热水溶解 ,经QAE -sephadex -A - 2 5柱层析 ,得SP - 1、SP - 2和SP - 3三个多糖组分。SP - 2经sephadexG - 10 0柱层析及醋酸纤维薄膜电泳证明为均一组分 ;经高效凝胶色谱法测定其相对分子质量为 8 82× 10 4 ;将多糖组分SP - 2水解后 ,用糖腈乙酸酯衍生物气相色谱法分析鉴定得单一葡萄糖 ,红外光谱显示SP - 2为 β D 吡喃糖苷键构成。生物活性实验表明 ,SP- 2的质量浓度分别为 0 5、5、2mg/L时 ,芥菜籽、鸡冠花籽、番茄种子的发芽率分别比对照组增长 14%、12 %、9%Nostoc Commune as raw material, after refluxing with methyl alcohol for 5 h to remove fatty materials, was extracted 3 times by water 98°C for 3.8 h. The combined raffinate was concentrated under vacuum, protein removed by proteolytic enzyme once and by Sevag method 7 times, and dialyzed by distilled water for 2 d. By adding 3 times volume of φ (CH3CH2OH) = 95% alcohol, polysaccharides were precipitated and freeze-dried. Then the dried mass was dissolved in boiling water, three-water-soluble polysaccharides (SP-1, SP-2 and SP-3) were obtained by QAE-sephadex-A-25 chromatography. SP - 2 was shown to be homogeneous by sephadex G - 100 chromatography and acetate film electrophoresis. Molecular weight of SP - 2 was estimated to be 8.82 × 104 by HPLC. After hydrolysis, SP - 2 was found by GC to consist of only one kind of monosaccharide, viz. glucose. Infrared spectrum indicated that SP - 2 has β-glycosidic linkage of D-glycopyranoside. Results of biological activity tests showed that at concentration of 0.5, 5 and 2 mg/L, SP - 2 increased the germination rate of the seeds of Brassica juncea, cockscomb and Lycopersicum esculentum by 14%, 12% and 9% respectively as compared with the controls.
