摘 要:为探讨补肾健脾固本法治疗哮喘的作用机理 ,将 72只雄性SD大鼠 ,随机分为正常对照组 (A)、哮喘模型组(B)、止喘胶囊小剂量组 (C)、止喘胶囊大剂量组 (D)、西药对照组 (E)和中西药结合组 (F) 6组 ,每组 12只。除A组外 ,采用腹腔注射卵白蛋白和灭活百日咳杆菌的方法 ,制成大鼠哮喘模型。在此基础上 ,各组模型鼠分别给予不同的药物“治疗” ,两周后处死测定各组大鼠血浆及支气管肺泡灌洗液 (BALF)中NO、ET -1和IL -5含量。结果 :B组BALF中NO浓度、ET -1含量明显高于A组 ;与B组相比 ,D、E组显著下降 ,后两组之间无统计学差异 ,F组BALF中NO浓度显著低于D组、E组 ,接近A组。B组血浆和BALF中IL -5含量均明显高于A组 ;与B组相比 ,除C组外 ,其余各治疗组均明显下降 ;其中F组显著低于D组、E组 ,接近A组。提示 :通过下调血浆和BALF中IL -5 ,抑制BALF中NO、ET -1的合成和释放 ,从而减轻哮喘气道炎症及气道上皮重建 ,降低气道高反应性 ,可能是补肾健脾固本法治疗哮喘的机制之一。In order to explore the functional mechanism of the kidney-tonifying,spleen-strengthening and constitution-consolidating method in the treatment of brachial asthma,72 male SD rats were divided randomly into six groups of the control group(A),asthma model group(B),small dose group of 'Wheezing-Stopping Capsules'(C),large dose group of'Wheezing-Stopping Capsules'(D),medication control group(E),and TCM and WIM-integrated group(F),12 rats in each group. Beside Group A,egg albumin was injected into the abdominal cavity plus the method to kill Bordetella pertussis,to create asthma model in the rats.Then,the model rats in various groups were given different medicines respectively.Two weeks later,the rats were killed to determine NO,ET-1 and IL-5 contents in plasma and bronchial alveolar lavage fluid(BALF)in the rats of various groups.The results showed that NO concentration and ET-1 content in BALF were obviously higher in Group B than in Group A.In comparison with Group B,the indexes remarkably dropped in Group D and E,but without statistical difference in the latter two groups,NO concentration in BALF was obviously lower in Group F than in Group D and E,closing to Group A.Plasma and IL-Content in BALF were obviously higher in Group B than in Group A.In comparison with Group B,except Group C,these indexes remarkably dropped in the rest groups,in which Group F was obviously lower than in Group D and E,colsing to Group A.The findings indicate that to relieve inflammation in the airway,reconstruct the epithelium of the airway in asthma and decrease high allergy in the airway by reducing plasma and IL-5 in BALF and inhibiting the synthesis and discharge of NO and ET-1 in BALF can be one of the mechanism in the treatment of asthma by the kidney-tonifying,spleen-strengthening and constitution-consolidating method.
