摘 要:目的 研究慢性肾功能衰竭 (CRF)患者低密度脂蛋白 (LDL)亚组份颗粒直径及氧化易感性的异常特征 ,探讨其动脉硬化高发生率的机制。方法 超速离心分离 30例CRF患者及 2 6例对照组的血浆LDL ,采用 2 %~ 16 %聚丙烯酰胺凝胶梯度电泳法分析LDL颗粒直径及图形并分析其抗氧化能力。结果 (1)CRF患者LDL颗粒直径 (2 6 .35±0 .5 9)nm同对照组 (2 6 .4 1± 0 .95 )nm间无差别 ,两组间A型、B型LDL图形分布亦相同 ;(2 )LDL颗粒直径同血浆TG水平负相关 (r =- 0 .2 6 ,P <0 .0 5 ) ;(3)CRF患者LDL抗氧化能力下降。Aim To study the size and susceptibility of low density lipoprotein (LDL) particles in patients with chronic renal failure (CRF). Methods Plasma LDL in 30 CRF patients and 26 control subjects were analyzed by 2%~16% non denaturing polyacrylamide gradient gels. The continuous monitoring of LDL oxidation by absorbance measurement at 234 nm was performed to analyze the susceptibility of LDL to Cu 2+ mediated oxidation in CRF patients. Results In LDL particle diameters no difference was found between the CRF patients and control subjects[( 26.35 ± 0.59) nm [WTBX]vs ( 26.41 ± 0.95) nm , P> 0.05 ]. The distribution of LDL subfraction pattern also had no difference between two groups. LDL particle diameter was negatively correlated with plasma triglycerides (. However, the lag time in the test for LDL oxidizabilty in CRF patients was significantly shorter than that in the controls[( 56.2 ± 23.7) min [WTBX]vs ( 97.3 ± 41.2 )min, P< 0.05 ]. . Conclusion Circulating LDL in CRF patients was more susceptible to oxidation in vitro than that of controls. This may be associated with increased risk for coronary artery disease.
