摘 要:在 30L搅拌式反应器中用多孔微载体培养分泌尿激酶原的DNA重组中国仓鼠卵巢细胞 (rCHO) ,研究了血清浓度及溶氧对细胞生长和表达的影响。结果表明 ,在 2× 10 5 ml低密度接种条件下 ,使用低 (无 )血清培养基会延长细胞生长延滞期并降低比生长速率 ,而细胞密度大于 10 6 ml时 ,血清浓度对细胞生长和产物表达的影响没有显著差别。溶氧 (DO)维持在 2 0 %~ 45 %时 ,对细胞表达产物和葡萄糖代谢无明显影响 ,但溶氧降至 7%~ 9%时 ,细胞表达水平明显降低 。A recombinant Chinese hamster ovary cell line, which secrets prourokinase (Pro-UK), was cultivated on porous microcarriers in a 30L stirred tank reactor. Cell growth and expression were studied in different serum concentration and dissolved oxygen (DO) level. The specific growth rate decreased and lag phase extended when cell grew in low serum or serum-free medium at low inoculated density of ~2.0×10 5/ml, but no significant effect on cell growth and expression was observed when cell density was more than 1.0×10 6/ml. There was few difference in cell expression and glucose metabolism when DO varied from 20%~45%. But when DO level dropped to 7%~9%, the cell expression level decreased and specific lactate production rate increased greatly.
