白细胞介素-10对鼠肾小球系膜细胞增殖和核因子κB的抑制作用

摘　　要：目的 :研究白细胞介素 - 10 (rhIL - 10 )对小鼠肾小球系膜细胞 (mesangialcell,MC)增殖的影响及其细胞内作用机制。方法 :采用 3H -TdR掺入法观察MC的增殖反应 ,采用电泳适移率改变法 (EMSA)检测核因子κB(nuclearfactorκB ,NF -κB)活性。结果 :rhIL - 10 (2 0ng/ml)抑制LPS刺激的小鼠MC增殖 (P <0 .0 5 ) ,同时rhIL - 10亦显著抑制LPS诱导的MC中NF -κB活化。结论 :IL - 10对LPS诱导的小鼠MC增殖的抑制作用至少部分与其对NFObjective: To investigate the effect of rhIL-10 on LPS-induced proliferation of mice mesangial cells and its relation with activation of nuclear factor κB. Methods: Mesangial cell proliferation was tested by 3H-thymidine uptake and activity of nuclear factor κB was measured by electrophoretic mobility shift assay. Results: rhIL-10(20 ng/ml) inhibited LPS-induced proliferation(P<0.05) and the activation of nuclear factor κB in mesangial cells induced by LPS. Conclusion: IL-10 can inhibit LPS-induced mesangial cell proliferation,probably mediated by decreased activation of nuclear factor κB.
