中药止喘胶囊对哮喘大鼠糖皮质激素受体及皮质酮的影响

摘　　要：目的 观察中药止喘胶囊对哮喘大鼠外周血白细胞和肺组织中糖皮质激素受体（ＧＣＲ）、血浆皮质酮、血浆及肺泡灌洗液（ＢＡＬＦ）中白细胞介素－４，５（ＩＬ－４，５）的影响，以探讨补肾健脾固本法治疗哮喘的作用机制。方法 采用放射性配基结合法、放射免疫法、酶联免疫法测定各组大鼠外周血白细胞和肺组织中 ＧＣＲ、血浆皮质酮、血浆及 ＢＡＬＦ中 ＩＬ－４，５含量。结果 哮喘模型组血浆皮质酮含量、外周血白细胞ＧＣＲ结合位点和肺组织 ＧＣＲ的 ＲＴ值均明显低于正常组（均Ｐ＜０．０１），中药止喘胶囊组血浆皮质酮含量、肺组织ＧＣＲ的ＲＴ值显著高于正常组（Ｐ＜０．０５，Ｐ＜０.０１）；哮喘模型组血浆和 ＢＡＬＦ中Ｌ－４，ＩＬ－５含量均明显高于正常组（均 Ｐ＜０．０１），其余各治疗组均明显下降（均 Ｐ＜０．０１），其中中西药结合组显著低于中药组、西药组（均Ｐ＜０．０１），接近正常组（Ｐ＞０．０５）。结论 止喘胶囊以补肾健脾固本治法通过阻止哮喘大鼠外周血白细胞和肺组织ＧＣＲ结合位点的下降，上调内源性皮质酮水平，改善下丘脑－垂体－肾上腺功能，下调血浆和 ＢＡＬＦ中 ＩＬ－４，５，从而减轻哮喘气道炎症，止喘胶囊与吸入激素有一定程度的协同作用。Objective To observe the effect of ZhiChuan capsule on glucocorticoids receptor and corticosterone in asthmatic rat and discuss the mechanism of BuShenJianPiGuBen method of traditional Chinese medicine in treatment of asthma. Methods The levels of glucocorticoids receptor( GCR) , corticosterone and interleukin - 4, 5 (IL - 4, 5) from rat were detected by immunolabelling technique. Results The levels of conjugated site and RT of GCR, corticosterone in model group were lower than that in normal group( P <0.01), but the levels RT of GCR, corticosterone in ZhiChuan capsule - treated group were higher than that in normal group ( P < 0.01 , P <0.05). The levels of IL- 4,5 in ZhiChuan capsule plus glucocorticoids group were significantly lower than that in group of ZhiChuan capsule - treated or glucocorticoids - treated (P <0.01) , and no differences between group of ZhiChuan capsule plus glucocorticoids - treated and normal (P > 0.05). Conclusion One of the mechanism of BuShenJianPiGuBen method in treatment of asthma may be alleviating airway inflammation through blocking down levels of GCR, increasing levels of endogenic corticosterone and reducing levels of IL -4,5. ZhiChuan capsule have part synergy with inhaled glucocorticoids.
