摘  要:目的检测云南恶性疟原虫多药抗性基因 (Pfmdr1 ) Asn 86Tyr的多态性 ,探索其与氯喹敏感性表现型的关系。方法用等位基因特异 PCR和限制性酶切片段长度分析技术 (AS-PCR/RFLP)。结果在 86% (1 9/2 2 )的氯喹抗性分离株中检测到 Pfmdr1基因的 Asn86Tyr两种多态性 ,携带编码 86Tyr多态变异的占 50 % ,而含编码 Asn86多态的也是 50 %。结论在云南氯喹抗性高度流行区 ,Pfmdr1基因Novel allele specific PCR and RFLP have been used to 22 isolates for detecting the mutation of Pfmdr1 Asn 86 Tyr associated with chloroquine resistance in Yunnan Province so as to explore an agreement between chloroquine resistance phenotype and the point mutation. The results showed that a total of 19 (86%) isolates detected out the Pfmdr1 Asn 86 Tyr , of which 10 chloroquine resistance isolates did not show the point mutation of the Pfmdr1 86 Tyr , while 9 isolates revealed the point mutation (A to T) of the Pfmdr1 86 Tyr . The incomplete agreement found between chloroquine resistance phenotype and Pfmdr1 (86 Tyr ) mutation would support the view that other genetic factors as well as Pfmdr1 may be involved in chloroquine resistance.
