摘 要:目的 为探讨移植物动脉硬化形成过程中 ,血管内膜和外膜增生的平滑肌细胞中是否有 Fas-配体 (Fas-L )表达。 方法 健康纯系 Wistar大白鼠为供者 ,30 6只 ;SD大白鼠为受者 ,16 2只 ,根据受者血管移植后有无明显排斥反应分为 3组。急性排斥组 :受者 90只 ,分别于术后 14天、2 8天、5 6天处死 ,每次处死 30只 ;免疫耐受组 :受者 72只 ,分别于术后 2 8天、5 6天、84天处死 ,每次处死 2 4只 ;对照组 :供者和受者均为 Wistar大白鼠 ,受者 72只 ,处死方法同免疫耐受组。采用同种异体大鼠胸主动脉腹腔移植模型。切取标本经处理后进行 Masson染色和免疫组织化学染色 ,比较移植术后不同时间 Fas- L 在增生外膜和内膜平滑肌细胞上的表达水平。 结果 术后 14天、2 8天、5 6天急性排斥组外膜 Fas- L的表达程度逐渐增高 (P<0 .0 1) ,其在外膜增生的平滑肌细胞中表达比内膜更早且广泛。 Fas- L在急性排斥组的表达程度和范围亦比免疫耐受组和对照组明显增高和广泛 (P<0 .0 5 )。 结论 在移植物动脉硬化形成过程中 ,Fas- L在增生平滑肌细胞中高度表达 ,其参与移植物动脉硬化的发生和发展。Objective By means of the rat abdominal aortic allograft model, to investigate whether there is high expression of Fas L or not in proliferative smooth muscle cells (SMC) of intima and adventitia during allograft arteriosclerosis(AA) formation. Methods Inbred male Wistar rats (306) served as donors, and Spregue Dawley(SD) rats (162) as recipients . They were classified into three groups. Acute rejection group:90 recipients were killed 14,28 and 56 days after transplantation,30 rats each time; immune tolerence group:72 recipients were killed 28,56 and 84 days after transplantation,24 rats each time; control group: 72 recipients were killed 28,56 and 84 days after operation, 24 rats each time.The rat abdominal aortic allograft model was used. The specimens were treated first and the immunohistochemistry was then taken to compare the smooth muscle α actin, Fas L and proliferating cell nuclear antigen (PCNA)expression degree in proliferating SMC of intima and adventitia during different times after operation. Results Followed by AA developing and progressing,the Fas L expression in adventitia increased gradually in 14,28 and 56 days postoperatively in acute rejection group ( P <0.01), and that in adventitia were expressed earlier and more extensive than that in intima.Compared with immune tolerence group and control group, the Fas L expression degree in transplanted vessels in acute rejection group were much higher and more extensive ( P < 0.05). Conclusion Fas L is highly expressing in proliferating SMC during AA formation, and takes part in AA formation.
