摘 要:目的 研究盐酸芦氟沙星 (rufloxacin hydrochloride,简称 RUFX)的电化学行为和电极反应机理 ,拟定了测定 RUFX的吸附伏安法。方法 用单扫示波极谱法、循环伏安法和直流极谱法等多种技术进行研究。结果 在 KH2 PO4- Na2 HPO4(p H7.0 0 )底液中 ,RUFX在汞电极上有一线性扫描还原峰 ,峰电位 Ep=- 1.2 7V(vs.Ag/Ag Cl) ,该峰具有明显的吸附性。吸附粒子为 RUFX中性分子 ,测得 RUFX在汞电极上的饱和吸附量为 5 .5 1× 10 - 1 1 mol/cm2 ,每个 RUFX分子所占电极面积为 3.0 1nm2 ,RU FX在悬汞电极上的吸附符合 Frumkin等温式。测得吸附系数 β=7.74× 10 5,电子转移数 (n)为 2 ,不可逆吸附的电子转移系数 (α)为0 .76 ,表面电极反应的速率常数 ks=0 .2 3/s。建立了吸附伏安法测定 RUFX的最佳条件 ,检出限为 1.0× 10 - 9mol/L。结论 证实该体系为具有吸附性的不可逆过程。AIM: To study the electrochemical behavior and reaction mechanism of rufloxacin hydrochloride (RUFX) in phosphate buffer solution. A method of Adsorptive Stripping Voltammetry for the determination of RUFX was established. METHODS: Linear Sweep polarography, cyclic Voltammetry and direct current polarography were used in the present research. RESULTS: In a supporting electrolyte containing KH 2PO 4-Na 2HPO 4 (pH7.00), a reduction peak of RUFX was observed by linear sweep voltammetry at Hg electrode. The peak shows a potential of -1.27V(vs.Ag/AgCl) and adsorptive characteristics. The adsorbed species is most probably the neutral molecular of RUFX. The saturated adsorption amount of RUFX at Hg electrode is 5.51×10 -11 mol/cm 2 and every RUFX molecule occupies an area of 3.01nm 2. On the surface of the hanging mercury drop electrode, the adsorption of RUFX obeys Frumkin adsorption isotherm. The adsorption coefficient β is 7.74×10 5, the number of electrons transferred (n) is 2, the transfer coefficient α is 0.76, the rate constant of surface electrode reaction k s is 0.23s -1 . The mechanism of electrochemical reduction of RUFX at Hg electrode is explored. The detection limit of the adsorptive stripping voltammetry of RUFX is 1.0×10 -9 mol/L under optimized condition. CONCLUSION: This indicates an irreversible process with adsorption.
