摘  要:目的研究雌激素替代治疗(estrogenreplacementtherapy,ERT)对去卵巢骨质疏松(ovariectomizedosteoporosis,OVX-OP)大鼠腺垂体滤泡星形(folliculo-stellate,FS)细胞中神经组织蛋白S-100、间隙连接蛋白Cx43表达的影响。方法采用10月龄、未孕产SD雌性大鼠60只,随机分为卵巢切除组(OVX)、假性手术对照组(Sham)和卵巢切除+雌激素替代治疗组(OVX+ERT),每组20只。于术后6周末应用双能X线骨吸收测量法(DEXA)检测Sham组和OVX组大鼠L4~6骨密度(bonemin-eraldensity,BMD)。OVX+ERT组大鼠于OVX术后第7~12周按照每周1mg/kg体重的剂量给予尼尔雌醇,于术后12周末检测大鼠L4~6BMD。制备各组大鼠腺垂体标本切片,应用间接免疫荧光法结合激光扫描共聚焦显微镜(LSCM)定位和定量分析腺垂体FS细胞中S-100、Cx43的表达。结果术后6周末Sham组L4~6BMD(0.211±0.013)g/cm2及术后12周末的OVX+ERT组L4~6BMD(0.204±0.010)g/cm2均明显高于术后6周末OVX组L4~6BMD(0.193±0.006)g/cm2(P<0.01)。S-100蛋白定位于FS细胞的胞浆中,Sham组、OVX组和OVX+ERT组中S-100蛋白表达的阳性率分别为80%、85%和85%,差异无显著性意义(P>0.05)。Cx43蛋白免疫荧光阳性信号定位于相邻的FS细胞以及相邻的FS细胞与内分泌细胞的胞浆中和(或)胞膜上。Objective To investigate the expression of nerve tissue protein S-100(S-100)and gap junctional protein connexin 43(Cx43) in rat adenohypophyseal folliculo stellate cells(FSCs) under the influence of ovariectomized osteoporosis(OVX-OP) and estrogen replacement therapy(ERT). Methods Sixty 10 month old female rats were randomly divided into sham operation control group(Sham), ovariectomy group (OVX) and ovariectomy+ERT group (OVX+ERT). L4-6 bone mineral density(BMD) of the animals in Sham group and OVX group were measured by dual energy X-ray absorptiometry(DEXA) at the end of 6 weeks after operation. From 7th week, rats in OVX+ERT group were treated with nilestriol(1 mg·kg -1·week-1) for 6 weeks. Rat L4-6 BMD in OVX+ERT group were measured at the end of 12 weeks after operation. Expression of S-100 and Cx43 in FSCs was detected using immunofluorescent histochemistry and laser scanning confocal microscopy(LSCM). Results L4-6 BMD was significantly lower in OVX group (0.193±0.006) g/cm2 than in Sham group(0.211±0.013) g/cm2 and in OVX+ERT group (0.204±0.010) g/cm2 (P< 0.01). No discernable difference was seen in the 3 groups on the fluorescence intensity and positive rate of FITC conjugated S-100 in FSCs . Whereas, Cx43 positive cell rate was markedly lower in OVX group(25%) than in Sham group(90%) and in OVX+ERT group(75%)(P< 0.005). Conclusion Low BMD provided definite evidence for osteoporosis induced by ovariectomy, and nilestriol effectively increase L4-6 BMD in ovariectomized rats. The stability of S-100 expression indicated that the number of FSCs had no correlation with the occurrence of OVX-OP and the therapeutic effect of ERT, whereas the decrease of Cx43 protein expression in the FSCs of the OVX and OVX+ERT groups and relatively augmentation were respectively closely related with the osteoporosis induced by estrogen deficiency and the therapeutic effect of ERT.
