摘 要:目的:测定不同甲状腺功能状态时大鼠24 h尿样中异喹胍(DB)和其体内主要代谢产物4-羟异喹 胍(4-OH-DB)的含量及其代谢比值(MR)。方法:采用气相色谱技术。结果:对照组、甲亢组和甲低组大鼠灌胃DB 0.5 mg·kg-1,求得其MR比值分别为: 0. 164±0.04,0.084±0.011和0.323±0.046(P<0.01) ; 其24 h尿中DB含量(μmol·l-1)分别为:64.8±5.5,48.4±3.0 和100.2±12.9(P<0.01) ;24 h尿中4-OH-DB含量(μmol·l-1)分 别为:416.7±86.6,574.3±72.0(P<0.01)和314.6±39.6(P <0.05)结论:甲状腺功能提高时大鼠对异喹胍的代谢能力明显增加,消除加速;甲状腺功能降低则相反。OBJECTIVE To determine the concentration of debrisoquine (DB)and its essential metabolite 4-hydroxy debrisoquine (4-OHDB) in rat's 24-hours urine on the different thyr oid functional state,and the ratio between debrisoquine and 4-hydroxy debrisoqu ine (metabolic ratio,MR)in urine. METHODS Gas chromatography was used with flame ionization detector(GC-FID). RESULTS The metabolic ratio (MR)of debrisoquine following ip 0.5 mg·kg-1 in c ontrol, hyperthyroidism and hypothyroidism group rats were:0.164±0.04,0 .084±0.011,0.323±0.046(P<0.01)respectively;The concentration o f debrisoquine in 24-hours urine were (μmol·l-1):64.8±5.5,48.4 ±3.0(P<0.01),100.2±12.90 (P<0.01) ;The concentration 7 of 4-hydrovy debrisoquine in 24-hours urine were (μmol·l-1):416.6± 86.6,574.3±72.0(P<0.01,314.6±39.6(P<0.05. CONCLUSIONS Debrisoquine hydroxylation increased on the hyperthyroidism rats.
