摘  要:目的 研究牛磺酸、茵陈素对顺铂引起的原代培养肾小管上皮细胞 (PTC)内游离Ca2 +浓度变化的影响。方法 在体外培养PTC、顺铂与PTC共同保温 2 4h ,牛磺酸、茵陈素分别提前与PTC作用 2 4h ,然后再加入顺铂 ( 2 6 μmol·L- 1 )共同保温 2 4h。采用Fura 2 AM测细胞内游离Ca2 +浓度。结果 顺铂 ( 13,2 6 ,5 2 μmol·L- 1 )升高PTC细胞内游离Ca2 +浓度 ,牛磺酸 ( 1,10g·L- 1 )和茵陈素 ( 4 ,8mg·L- 1 ) ,可拮抗顺铂导致的PTC细胞内游离Ca2 +浓度超载。结论 顺铂可引起原代培养肾小管上皮细胞内Ca2 +超载 ,牛磺酸、茵陈素能拮抗顺铂导致的细胞内Ca2 +超载 ,起到细胞保护作用。Objective To study cisplatin induced cytosolic free [Ca 2+ ]i overload in primary culture of proximal tubular cells (PTC), and the preventing effect of taurine and scoparoneon in this event. Methods After proximal tubular cells (PTC) were isolated from New Zealand rabbits and cultured, some of them were incubated with cisplatin (0, 6.5, 13, 26, 52 μmol/L) for 24 h to induce cytosolic free [Ca 2+ ]. And others were cultured in the mediums with taurine or scoparoneon for 24 h, and then for another 24 h after cisplatin (26 μmol/L) addition. Cytosolic free [Ca 2+ ]i overload was measured with Fura 2/AM. Results ① Cisplatin (13, 26, 52 μmol/L) increased the concentration of Ca 2+ in PTC;②Taurine (1, 10 g/L) and scoparoneon (4, 8 mg/L) significantly decreased cisplatin induced cytosolic free i overload respectively. Conclusion Cisplatin can induce cytosolic free [Ca 2+ ]i overload in PTC, which can be prevented by taurine and scoparoneon.
