摘  要:目的 观察肝硬化大鼠肾脏水通道蛋白 2 (AQP2 )和血管紧张素Ⅱ -Ⅰ型受体 (AT1)受体、转化生长因子 β(TGFβ)的表达情况 ,及中药黄芪的治疗作用。 方法 肾脏皮髓质AQP2mRNA检测采用半定量RT PCR法 ,肾脏AT1受体、TGFβ蛋白检测采用免疫组化法。 结果 肝硬化 2周和 5周大鼠肾脏皮髓质AQP2mRNA和AT1受体、TGFβ蛋白表达均上调 ,黄芪在 2周时可纠正这些指标的升高 ,5周时则只能降低AT1受体的高表达。结论 肝硬化大鼠存在AQP2表达的异常 ,黄芪在早期具有改善作用。Purpose: To investigate the changes of AQP2, AT 1 receptor and TGFβ in liver cirrhosis and the role of astragalus memberanaceus(AM). Methods: Immunohistochemistry and relatively quantitative RT-PCR technique were used to detetmine the renal protein levels of AT 1 receptor and TGFβ and mRNA expression of AQP2 in cirrhotic rats induced by common bile duct ligation (BDL) for 2 and 5 weeks. Results: The mRNA expression of AQP2 and protein expression of AT 1 receptor and TGFβ were up regulated in the kidney of cirrhotic rats. AM could alleviate the over expression of these parameters in compensated cirrhosis (2 weeks), but have no effect on decompensated cirrhosis (5 weeks). Conclusions: The abnormality of AQP2 exists in cirrhosis, AM has a therapeutic effect on compensated cirrhosis. Its mechanism amy be through the inhibition of RAS.
