摘  要:目的 :研究先天性HCMV感染与胎鼠大脑皮质异常的关系。方法 :10周龄的ICR雌雄鼠按 1∶1配对 ,妊 7天时实验组行腹腔内接种HCMV ,对照组则接种HF细胞上清液。分别于妊 12、15、19天剖腹取胚胎大脑皮质进行病理学检查、病毒分离及原位杂交。结果 :实验组ICR胎鼠脑实质血管充血、出血等炎性改变 ,妊 19天胎鼠大脑皮质神经细胞变性坏死 ;3个时间点实验组病毒分离率分别为 10 %、40 %及 6 7% ,MIE和LA寡核苷酸探针原位杂交阳性率为 30 %、6 0 %、83%和 0 %、40 %、70 %。结论 :HCMV感染妊 7天ICR鼠后 ,不仅可在胎鼠大脑皮质神经细胞中复制且使大脑皮质损伤明显。In order to research effect of congenital human cytomegalovirus (HCMV) infection on the disorder of cerebral cortex, HCMV (6.0logTCID 50 In1.0 ml/mice) was injected into the intraperitoneum of 10-week-old female mice on 7 gestation days. The cerebral cortex was removed from the skull aseptically on day 12,15,19 of gestation, and then stained with HE. Meanwhile, part of the specimens was used for viral isolation and HCMV DNA detection by in situ hybridization. Pathological changes in the fetal cerebral cortex showed that the capillary was congested and bled, and the degeneration and necrosis of neurons co-existed on 19 th gestation day. The positive rate of virus isolation of the fetal cerebral cortex at day 12,15,19 of gestation was 10%, 40%, 67%. The positive rate in situ hybridization using MIE and LA probes were 30%, 40%, 83% and 0%, 40%,70% respectively. The fetal cerebral cortex can not only be replicated but damaged by HCMV on experiment..
