卵母细胞核移植中去核技术的探讨

摘　　要：目的 :了解卵母细胞进行去核手术时其去核方法的可靠性。方法 :通过 2步法对 175 6枚卵母细胞进行去核操作。结果 :存活 12 46枚 ,存活率 71 1% ,将存活卵子进行固定、染色后观察 ,去核完全的卵母细胞有 10 70枚 ,去核率为85 9% ,总去核率为 6 1 0 %。结论 :此法去核有一定可靠性。细胞核的定位是手术成功的关键 ,固定针、去核针。Objective:To study the reliability of enucleation of recipient oocytes through nuclear transplantation.Methods:A simplified method was used here for mouse enucleation-TWO STEPS.Results:1 756 recipient oocytes were enucleated,1 246 survived.the survival rate was 71 1%,the survival oocytes stained.there were 1 070 oocytes without nuclear.the enucleated rate was 85 9% and The total enucleated rate was 61 0%.Conclution:The method of enucleation was reliable.The location of the nuclear was the key to the operation,while the preparation of the injected needles and enucleated needle was indispensable in nuclear transplantation.
