摘 要:目的 :观察五子衍宗丸及其拆方对老年线粒体 DNA缺失、线粒体呼吸链酶复合体及三磷酸腺苷(ATP)合成的影响 ,以探讨其延缓衰老的机制。方法 :38例老年肾虚健康男性 ,随机分为五子衍宗丸治疗组和安慰剂组 ,服药 3个月 ,研究采用单盲法。线粒体 DNA缺失、线粒体呼吸链酶复合体的活力及 ATP合成分别用聚合酶链反应 (PCR)、酶动力学和生物发光技术检测。结果 :临床研究发现 :五子衍宗丸可提高老年男性外周血白细胞线粒体呼吸链酶复合体 、 活力 ,减少线粒体 DNA缺失。动物实验发现 :五子衍宗丸及其拆方的枸杞子、菟丝子可减少老年大鼠脑组织线粒体 DNA缺失 ,提高鼠脑线粒体呼吸链复合酶 、 活力和脑线粒体ATP的合成。结论 :五子衍宗丸对老年人及老年大鼠线粒体 DNA氧化损伤有保护作用。Objective:To investigate the effect of Wuzi Yanzong pill(WZYZP,五字衍宗丸) and its disassembled prescription on mitochondrial DNA(mtDNA) deletions,respiratory chain complexes and ATP synthesis.Methods:Thirtyeight aged men with symptoms of senility due to deficiency syndrome of the kidney,but otherwise healthy,were randomly divided into two groups:one group was treated with WZYZP and the other with a placebo,each for three months,a singleblind method was used.mtDNA deletions,respiratory chain complexes and ATP synthesis were determined with polymerase chain reaction(PCR),enzyme kinetics and bioluminescence technique respectively.Results:WZYZP could reduce the mtDNA deletions and raise the activities of mitochondrial respiratory chain complexes Ⅰ,Ⅳ in aged men of periphery blood leukocytes.WZYZP and its disassembled prescription of Fructus Lycii(Wolfberry Fruit,枸杞子) and Semen Cusuctae(Dodder Seed,菟丝子)could reduce the mitochondrial DNA deletions and raise the activities of mitochondrial respiratory chain complexes Ⅰ,Ⅳ and ATP synthesis in aged rat brain.Conclusions:WZYZP have protective effect on oxidative damage of mtDNA in aged men and aged rats.
