降钙素基因相关肽对油酸型急性肺损伤家兔细胞因子的影响

摘　　要：目的　探讨降钙素基因相关肽 (CGRP)对油酸型急性肺损伤 (acute lung injury,AL I)家兔血与支气管肺泡灌洗液 (BAL F)中 TNF- α,IL- 8和 ET- 1含量的影响 .方法　观察油酸型 AL I家兔 CGRP治疗前、后不同时间静脉血与 BAL F中上述 3种细胞因子的含量 ,并测定其肺含水量 .结果　油酸致伤后 30 ,6 0 ,12 0 min血及 BAL F中 TNF-α,IL - 8和 ET- 1含量均显著增高 (P<0 .0 5～ 0 .0 1) ,尤其以 TNF-α增幅最大 .CGRP治疗后各因子含量明显下降 (P<0 .0 5 ) ,并维持在低水平 ,肺含水量也显著降低 (P<0 .0 5 ) .结论　静脉注射 CGRP可有效降低油酸致伤后血及 BAL F中 TNF- α,IL- 8和 ET- 1水平 ,抑制肺内炎症反应 。AIM To investigate the effects of calcitonin gene-related peptide (CGRP) on the levels of TNF-α, IL-8 and ET-1 in blood and br onchio-alveolar lavage fluid (BALF) from rabbits with acute lung injuries (ALI) induced by oleic acid. METHODS Concentrations of the three cytokines in venous blood and BALF were continuously analyzed at different time before and after the treatment with CGRP (0, 30, 60, 120 min). Additionally, extravascular lung water was measured at the end of the study. RESULTS Concentrations of th e three cytokines in blood and BALF were markedly increasd at 30, 60 and 120 min after intravenous infusion of oleic acid (P<0.05~0.01). In contrast, the levels of the three cytokines remarkably decreased and remained in a lower state after the intravenous infusion of CGRP (P<0.05). So did the extravascular lung water (P<0.05). CONCLUSION The intra venous infusion of CGRP may efficiently decrease the levels of TNF-α, IL-8 and ET-1 in blood and BALF after the infusion of oleic acid, inhibit intrapulmonary inflammation and reduce the lung injury.
