摘　　要：目的　探讨IL 1β、IFN γ对基质金属蛋白酶 - 2 ,- 9(MMP - 2 ,- 9)mRNA基因表达的影响。 方法　采用体外灌流卵巢系统、原位杂交 (insituhybridization)等方法检测IL 1β、IFN γ对大鼠卵泡发育过程中MMP - 2 ,- 9mRNA基因表达变化 ,并通过密度扫描方法对原位杂交结果进行半定量分析。结果　IL 1β对MMP - 2mRNA的表达无明显的刺激或抑制作用 ,但可显著增加卵泡膜细胞的MMP - 9mRNA的表达 ;IFN -γ可显著降低MMP - 2 ,- 9mRNA的表达。结论　IL 1β、IFN γ可通过调节卵泡发育过程中MMP - 2 ,-Objective To study the effects of IL 1β and IFN γ on expressions of matrix metalloproteinases-2,-9(MMP-2,-9) mRNA in the rat ovary. Methods MMP-2,-9 mRNA expressions during follicular development in the rat ovary were assessed by in situ hybridization and in vitro -perfusion ovary system, and the results were determined by semi-quantitative analysis through densitive scanning method.Results IL 1β had no stimulated or inhibited effect on expression of MMP-2mRNA, but the expression of MMP-9 mRNA in theca cells(TC) was apparently increased by IL-1β. The expressions of MMP-2,-9 mRNA were apparently decreased by IFN-γ.Conclusion IL-1β and IFN-γ might regulate the follicular development through regulated MMP-2,-9 gene expressing changes during the follicular development.
