摘  要:目的 研究自体骨骼肌卫星细胞移植对梗死心肌残存心肌细胞体积的影响。方法 自犬臀部取骨骼肌 ,体外提取、培养、扩增骨骼肌卫星细胞 ,4’ ,6 diamidino 2 phenylindone(DAPI)荧光标记 ,经结扎的左冠状动脉前降支远端灌注移植入自体急性心肌梗死区域。 2、4、8周后处死动物 ,取心肌标本行组织形态学及心肌细胞体积检测。结果 梗死区部位观察到带荧光的卫星细胞及肌纤维 ,部分已分化成横纹肌并以闰盘相连。正常心肌细胞随梗死时间增长 ,可见细胞切面积及体积增加 ,呈现肥大趋势。骨骼肌卫星细胞移植治疗组梗死区残存心肌细胞肥大明显 ,细胞切面积、面积、体积与对照组比较差异有显著意义。细胞肥大以细胞宽度及厚度为主 ,细胞长度未见明显变化。结论 骨骼肌卫星细胞移植入梗死心肌 ,可促使梗死区残存心肌细胞肥大 。Objective: In this experimental study, the effect of skeletal muscle satellite cell implanted into infarcted myocardium on remnant myocytes volumes was studied. Methods: Canine skeletal muscle satellite cells were procured, cultured, proliferated and then labeled with 4',6 diamidino 2 phenylindone(DAPI) in vitro. The cells were autologously implanted into the site of acute myocardial infarction by infusion through distal portion of ligated left anterior descending coronary artery and the specimens were harvested 2, 4, 8 weeks later for histologic study and the examination for myocytes volume. Results: The satellite cells and myofiber with fluorescent were observed in the infarct site. A portion of the implanted cells had differentiated into muscle fiber with striation and connected with intercalated discs. Hypertrophy of remnant myocytes cells in infarcted site after skeletal muscle cell implantation was evident. The cross sectional area, cell area and cell volume was remarkably different from that of control. The hypertrophy of cell was in width and thickness, and the length remained unchanged. Conclusion: The skeletal muscle satellite cells, implanted into infarct myocardium, could induce the hypertrophy of remnant myocyts cell in infarcted site and may also favor the recovery of contractile force.
