高效液相色谱法同时测定心律平及乙吗噻嗪血药浓度

摘　　要：心律平(Propafenone,PPF)为I_c类抗心律失常药,乙吗噻嗪(Ethmozine,EMZ)为I_b类抗心律失常药,两药国内均于1985年投入市场。近年来研究表明两者合用对难治性心律失常有协同作用,但个体差异大,生物利用度较低。A simple and sensitive high performance liquid chromatographic(HPLC) method was established to assay propafenone and ethmozine concentrations in human plasma simultaneously. The sample,internal standard and n-heptane/isomylalcohol(99: 1,V/V)were added into a tube and the mixture was snaked and centrifuged. After frozen, the supernatant was transferred and H3FO4 was added. A portion of the acid phase was then injected into the HPLC. Separation was obtained using ODS column and methanol- buffer solution- acetonitrile- diethylamine (50 : 29. 5 : 20 : 0. 5,V/V)mobile phase. The column effluent was monitored at 254nm. For both drugs, the recoveries >95%,the CV≤7. 17% for within-day and day-to-day the sensitivity = 10ng/ml and the method was linear in the range of 50- 2500ng/mk with γ>0. 998.
