摘 要:目的 探讨丹参对肝脏保存再灌注时Bcl 2、Bax表达和肝细胞凋亡的作用。方法 建立离体大鼠肝脏保存再灌注模型 ,分别采用流式细胞仪和原位末端标记 (TUNEL)技术 ,观察Bcl 2、Bax表达和肝细胞凋亡以及丹参对上述指标的影响。结果 丹参组保存 16、2 4、3 2h肝脏Bcl 2表达分别为 6 2 9± 0 0 5、4 88± 0 13及 4 2 7± 0 3 0 ,明显高于对照组 5 5 4± 0 4 4、4 3 2± 0 3 9及 3 79± 0 2 0(P <0 0 5 )。丹参组bax表达分别为 3 3 4± 0 0 8、4 74± 0 2 2及 5 2 2± 0 0 9,明显低于对照组 4 13±0 3 6、5 77± 0 3 9及 6 2 3± 0 11(P <0 0 5 )。丹参组肝细胞凋亡指数则分别为 4 7 6± 1 4、2 93± 12及5 3 8± 8明显低于对照组 81± 4、5 0 6± 7及 664± 12 (P <0 0 5 )。结论 丹参可使肝脏保存再灌注后Bcl 2表达增强 ,Bax表达减弱 ,肝细胞凋亡减少。因而可以预防肝脏保存再灌注损伤。Objective To investigate the effect of salvia miltiorrhiza (SM) on the expression of Bcl 2?Bax and hepatocyte apoptosis in hepatic preservation and reperfusion in a murine model.Methods The model of the preservation reperfusion of isolated rat liver was established. Bcl 2?Bax expression of the liver tissue and hepatocyte apoptosis were studied respectively by flow cytometry and in situ terminal deoxynucleotidyl transferase mediated dUTP FITC nick end labeling (TUNEL) technique. Results In SM treated group levels of Bcl 2 expression was significantly increased ( P <0 05), Bax expression was decreased ( P <0 05), hepatocyte apoptosis index was remarkably lowered ( P <0 05), compared correspondingly with control group after preservation for 16, 24 and 32*!h . Conclusion SM can increase Bcl 2 expression, decrease Bax expression and inhibit hepatocyte apoptosis, SM may play a positive effect in preventing hepatic preservation reperfusion injury.
