摘  要:目的 探讨甲基对硫磷 (M160 5 )的易感性生物标志物。方法 检测M160 5作业工人 ( 71名 )及对照组 ( 5 0名 )的对氧磷酶 (PON)基因在 192位点和 5 5位点的多态性分布 ,分析各基因型与全血胆碱酯酶 (ChE)活力、免疫功能和内分泌水平的关系。结果 在全部 12 1名研究对象中 ,PON在 192位点和 5 5位点基因型均为多态性分布 ;基因型分布及等位基因频率性别之间差异无显著性 (P >0 .0 5 )。在 71位M160 5接触者中 ,PON基因192位点为谷氨酸 (Gln) /Gln的基因型者 ,其T淋巴细胞亚群中CD4、CD4/CD8分别为 4 1.3± 5 .3、1.5 6± 0 .15 ,比对照组 ( 4 9.0± 8.1、1.83± 0 .10 )和M160 5接触组精氨酸 (Arg) /Arg基因型者 ( 4 4.9± 4 .8、1.76± 0 .14 )明显降低 ,差异有显著性 (P <0 .0 5、P <0 .0 1)。PON基因 5 5位点不同基因型者M160 5的免疫毒性作用差异无显著性 (P >0 .0 5 )。结论 PON基因在 192位点Gln/Gln基因型可能是M160 5免疫毒性作用的易感性生物标志物之一。Objective To investigate the susceptibility biomarkers of methylparathion(M1605) toxicity. Methods T5'BZ]Paraoxonase(PON) genotypes were determined by restriction fragment length polymorphism(RFLP).The toxic effects of M1605 on exposed subjects were evaluated by measuring the activity of whole blood cholinesterase (bl-ChE),concentration of serum IgG,IgA,IgM,and hormones(follicle-stimulating hormone,luteinizine hormone,and testosterone),and detecting T lymphocyte subpopulations.The relationships between the individual's genotypes according to two certain polymorphic sites on paraoxonase gene,and the toxic effects of M1605 were also explored. Results The distribution of PON genotypes at codon 192 and 55 was polymorphic in 121 subjects,and was not significantly different between males and females.Among 71 subjects exposed to M1605,those who were of PON-192 Gln/Gln genotype had significantly less CD 4 subpopulation and CD 4/CD 8(41.3±5.3 and 1.56±0.15) than control(49.0±8.1 and 1.83± 0.10) and those who were of PON-192 Arg/Arg genotype(44.9±4.8 and 1.76±0.14),P<0.05,P<0.01,but there was no evidence of relation between PON-55 genotype and toxic effects of M1605.
