摘  要:目的 研究辛硫磷 (Pho)与氰戊菊酯 (Fen)对大鼠睾丸的联合毒性作用。方法 Pho与Fen分别选择两个剂量水平 ( 1/180LD50 和 1/2 0LD50 )并作混配 ;SD雄性大鼠分成 7个剂量组 ,1次 /d灌胃 ,5d/周 ,连续 60d ,组成 2个 2× 2析因设计实验 ,即Pho组 ( 8.2、73 .5mg/kg体重 ) ,Fen组 ( 3 .3、3 0 .0mg/kg体重 ) ,两个Pho +Fen组分别为 (Pho 8.2 +Fen 3 .3 )、(Pho 73 .5 +Fen 3 0 .0 )mg/kg体重 ,1个对照组。结果 析因分析显示 ,Pho +Fen联合染毒 ,在低剂量水平 ,对大鼠精子运动参数、精子生成量、睾丸酶活力和睾酮 (T)含量等无交互影响 ,联合作用为相加作用 ;在高剂量水平 ,对精子的运动参数如直线运动速度(VSL)、曲线运动速度 (VCL)、鞭打频率 (BCF)、直线性 (LIN)、前向性 (STR)等和精子生成量、睾丸γ 谷氨酰转移酶 (γ GT)及乳酸脱氢酶 (LDH)活力、T水平等存在交互影响 ,表现为拮抗作用 ,对酸性磷酸酶(ACP)、碱性磷酸酶 (ALP)及葡萄糖 6 磷酸脱氢酶 (G 6PDH)活力无交互影响 ,表现为相加作用。结论 Pho和Fen低剂量水平对大鼠睾丸的联合作用特征呈相加作用 ,高剂量水平呈拮抗作用或相加作用 ;Pho和Fen两个剂量水平的联合作用与单剂农药相比 ,未见有明显的增毒效应 ,但均能对大鼠睾丸产生毒性。Objective To study the combined toxic effect of phoxim(Pho) and fenvalerate(Fen) on male rat testis. Methods The adult male SD rats were randomly divided into 7 groups(including a control and six treated groups).Various daily doses of Pho(8.2,73.5 mg/kg),Fen(3.3, 30.0 mg/kg),Pho+Fen(8.2+3.3 mg/kg) and Pho+Fen(73.5+30.0 mg/kg) were given to rats by gastric gavage for 60 successive days.2×2 factorial analysis was used in the experiment. Results (1)No interaction was noted between Pho and Fen on sperm motility and production,the activity of testicular enzyme and testosterone level at low mixed doses,the joint action was characterized as an additive effect.(2)At higher concentrations there were interactions between Pho and Fen on sperm motion parameters(VCL,VSL,BCF,LIN and STR),sperm production,the activities of testicular enzymes(LDH, γ-GT) and testosterone level and the joint action was characterized by antagonistic effect,but no interaction was found on the activity of other testicular enzymes(ACP,ALP,G-6PDH),and the joint action exhibited additive effect. Conclusions The joint actions of Pho and Fen on the testis of male rats were related to dose levels of the mixture,that is additive effect exhibited at low dose and antagonistic or additive effect at higher dose.No remarkably increased toxic effect of the pesticide mixtures was found as compared with each pesticide alone though the toxicities can be produced on rat testis at both low and higher concentrations.
