摘 要:用 1日龄黄羽肉鸡研究了一氧化氮 ( NO)对接种 Eimeria acervulina雏鸡体液免疫水平的影响。试验设 3个对照组 (免疫对照组、感染对照组、空白对照组 )和 3个试验组 ( L-精氨酸组、L-氨基胍组、NG-亚硝基 -L-精氨酸甲基酯组 ) ,每组 8只鸡。免疫程序为 ,7日龄时初免 ,接种卵囊 3× 1 0 3 个 /只 ;1 4日龄时二免 ,接种卵囊 5× 1 0 3 个 /只 ;2 1日龄时三免 ,接种卵囊 1× 1 0 4 个 /只 ;2 8日龄时四免 ,接种卵囊 3× 1 0 6个 /只。感染对照组用于判断本试验所采用的免疫程序是否成功 ,该组鸡仅在 2 8日龄时感染卵囊 1次 ,剂量为 3× 1 0 6个 /只。 3个对照组从 7日龄开始直至扑杀当日连续口服生理盐水 0 .5 m L/次 ,2次 /d;3个试验组从 7日龄开始直至扑杀当日分别口服 5 g/L L-精氨酸溶液或 3g/L L -氨基胍溶液或 6 g/L NG-亚硝基 -L-精氨酸甲基酯溶液 ,0 .5 m L/次 ,2次 /d。 38日龄时扑杀全部试验鸡 ,取血清样本用 ELISA法测定抗体水平 ;取十二指肠样本进行肠道病变记分。试验结果表明 ,给免疫 E.acervulina的雏鸡每日口服 NO生成底物 L -精氨酸 ,可增加其血清抗体水平 ( P<0 .0 5 ) ;而口服诱导型 NO合成酶 ( i NOS)抑制剂氨基胍和原生型 NO合成酶 ( c NOS)抑制剂 NG-亚硝基 -L -精氨酸甲基酯 ,可明?Forty eight Huangyu broilers after being fed starter feed for one week from one-day- old,were divided into control group,infective group,inoculated group,L-Arg-treated group,L-AG- treated group,and L -NAME-treated group.Of 8chickens each were used to study the effectof NO on systemic antibody level of chickens inoculated with E.acervulina.Chickens in the latterfour group re- ceived firstinoculation at7-day-old,inoculated dose to be3× 1 0 3 oocysts per chicken,second inocula- tion at1 4-day-old,with 5× 1 0 3 oocysts per chicken,third inoculation at2 1 -day-old,with 1× 1 0 4 oocysts per chicken.Fourth inoculation at2 8-day-old,with 3× 1 0 6oocystsperchicken.Chickens in in- fective group were infected at 2 8-day-old,with 3× 1 0 6oocysts per chicken.While chickens in control group were given no oocysts.Chickens in L-Arg-treated group,L-AG-treated group,and L-NAME- treated group were given separately0 .5m L 5mg/ m L L-Arg,or3 mg/ m L L-AG,or6mg/ m L L- NAME orally from 7-day-old to 3 8-day-old,twice a day.All chickens in the experiment were slaugh- tered at 3 8-day old.Collected blood sample for determination of antibody by ELISA and duodenum sample for lesion scored.The results showed that the antibody level of chickens in L-Arg-treated group was higher than thatin inoculated group( P<0 .0 5) ,while that in L-AG-treated group and L- NAME-treated group was lower than that in inoculated group( P<0 .0 1 ) .L esion scores in L-Arg- treated group was similar to thatin inoculated group,while lesion scores in L -AG-treated group and L-NAME-treated group were higherthan thatin inoculated group( P<0 .0 1 ) .This resultshowed that the increasied NO can improvethe ability of immunity of chickensagainst E.acervulina,while the inhi- bition of the production of NO would disturb the immunized process in chickens.Because L-NAME can specially inhibit the activity of c NOS,and L -AG can specially inhibitthe activity of i NOS,the re- sults of this experimentindicated that NO synthesed by both kind of NOS,as a e
