摘 要:建立了肉鸡组织中盐酸沙拉沙星的 HPLC测定方法 ,并测定了肉鸡单次内服 (每 1 kg体重 1 0 mg)和连续 5 d混饮 (每 1 L饮水 5 0 mg)给药后组织中的沙拉沙星残留特征。组织样品用乙腈 -氨水匀浆 ,离心后取上清液用正己烷 -乙醚除脂 ,水相用磷酸酸化后作 HPLC检测。色谱柱为 Hypersil C18柱 ,紫外检测波长 2 80nm。流动相为乙腈 -2 %西丁基溴化铵 (体积比 1 0∶ 90 ) ,用磷酸调 p H值至 2 .5。肝脏、肾脏、肌肉中沙拉沙星的检测限为 0 .0 5 μg/ g,组织样品回收率均大于 90 %。鸡单次内服盐酸沙拉沙星后 ,2 4 h内各组织中均可检出药物 ,4 8h后肝脏、肾脏、肌肉中药物残留量均低于 0 .0 5 μg/ g;混饮停药后 ,6 h内肝脏、肾脏中可检出残留药物 ,1 2 h后 3种组织中残留量均低于 0 .0 5 μg/ g。结果表明 ,盐酸沙拉沙星在肉鸡组织中消除迅速 。A HPLC method wasdeveloped for the determination of tissue residues of sarafloxacin in broiler chickens. Afterselective extraction with mixture of acetonitrile and ammonia,the tissue samples were cleaned up by N-hexane and ether,and sarafloxacin was separated on a hypersil C18column using acetonitrile and2 % tetrabutyl ammonium bromide ( 1 0 / 90 ,V/ V) adjusted to p H2 .5 with phosphoric acid as the mobile phase,and quantitated by using ultraviolentdetector set at2 80 nm.The mean recoveries were95 .5 1 % ,95 .97% and94 .71 % for the sarafloxacin in the livers,kidneys andmuscles,and detection limitfor sarafloxacin in chicken tissues was at a level of0 .0 5 μg/g.After oral administration of a single dose of1 0 mg/kg,sarafloxacin could be detected in livers,kidneys and muscles2 4 h afterdosing,the residue levels were lower than the detection limit4 8h after administrations.After repeated doses of5 successive days drinking water administration at the concentration of5 0 mg/L,sarafloxacin could be detected in livers and kidneys6 h after the last dose,there were no residues higher than 0 .0 5μg/g 1 2 h after withdrawal of administrations.The results showed thatthe depletion of sarafloxacin in broilerchickens is rapid with a shortresidue time.Sarafloxacin would notbe accumulated af- ter multiple administration.
