摘  要:目的 建立一套肌肉细胞凋亡的检测方法,探讨不同的运动量与骨骼肌细胞凋亡的关系。方法 采用流式细胞术检测肌肉细胞内的[Ca]2+浓度、线粒体膜电位、DNA周期分析及原位末端标记技术(TUNEL)。结果 大鼠运动训练后各组肌细胞内[Ca]2+明显高于对照组(P<0.01),运动后 1周增高最显著,第 2周后开始降低,2周、3周与运动后1周比较仍有显著性差异(P<0.05);线粒体膜电位在运动训练后1周降低,第2周开始明显增高,与对照组相比有显著性差异(P<0.01)。第3周下降,第2周、3周与第1周比仍有显著性差异。运动后各组的DNA周期分析均出现一亚“G1”峰即凋亡峰,TUNEL显示运动后1周凋亡细胞比例最高,2周、3周后比例降低,但运动组与对照组比较都有显著性差异(P<0.01)。结论 不同的运动量可引起不同程度的细胞凋亡。本检测方法较为系统、全面,特异性强、灵敏度高。Objective To establish a method of measuring cell apoptosis, and to investigate the relationship between apoptosis and different exercise amount. Methods Determine the concentration of calcium in muscular cells and mitochondrial membrane po-tential, and analysis the DNA cycle with the technique of flow cytometry, and TUNEL change. Results Compared with control group, a pronounced rising of [Ca]i2+ level occured during following weeks one week group and the other two groups (P<0.05). The mitochondrial membrane potential decreased after one week (P<0.05), then increased markedly after two weeks (P< 0.01) and didn't change significantly after one week (P<0.05) compared with control. However, the mitochondrial membrane potential of after two and three weeks was lower than that after two and three weeks (P<0.05). DNA cycle analysis demonstrat-ed that sub-Gi namely apoptosis peak appeared in three groups after exercise. TUNEL showed that the apoptosis rate augmented after exercise compared with control rats (P<0.01), and the rate was the highest after one week. Conclusion Different amount of exercise may cause apoptosis increasing in different degrees. These methods are relatively systemic, much more specificity and sensitivity.
