HPLC测定活血合剂中丹参素、原儿茶醛的含量

摘　　要：目的 :建立RP HPLC法测定活血合剂中丹参素、原儿茶醛的含量。方法 :采用离心 微孔滤膜过滤的除杂方法制备供试品溶液 ,色谱柱为 :InertsilODS 3,4.6× 2 5 0mm 5 μm ;流动相为甲醇 0 .5 %HAC水溶液 (2 2∶78) ,检测波长为 2 80nm ,进行测定。结果 :方法的平均加样回收率 :丹参素为 96 .7% ,RSD =1.12 % (n =5 ) ;原儿茶醛为 97.0 % ,RSD =1.5 3(n =5 )。丹参素在 1.2 5～ 10 μg范围内 ;原儿茶醛在 0 .2 5～ 2 .0 μg范围内 ,线性关系良好。 结论 :方法简便、准确 ,重现性好 。Objective:To establish the method of determination of the contents of Danshensu and Protocatechuic aldehyde in Huoxue Mixture simultaneously by RP HPLC.Methods:The determination was carried out with Inertsil ODS column. The mobile phase consisted of methanol 0.5% HAc (22∶78) and UV detection wavelength was 280nm. Results:The average recovery of standard addition were 96.7%, RSD =1.12% ( n =5) for Denshensu, and 97.0 %, RSD =1.53% ( n =5) for Protocatechuic aldehyde, There was a good linear relationship between the absorption area value and the concentration in the range of 1.25～10μg for Deshensu and 0.25μg～2.0μg for Protocatechuic aldehyde.Conclusion: The method is simple, acurate, reproducible and can be used for content determination of Huoxue Mixture.
