摘  要:目的 :建立HPLC法同时测定血清中 4种苯二氮类药物硝西泮、艾司唑仑、阿普唑仑、地西泮的浓度。方法 :血清样品在碱性条件下用乙醚 (已去除过氧化物 )提取 ,4 0℃水浴蒸干 ,残渣用流动相溶解 ,进样分析。色谱条件为DiscoveryC18柱 5 μm (2 5 0mm× 4 6mm) ,流动相为甲醇∶乙腈∶水 (42∶18∶4 0V/V ) ,检测波长2 30nm ,柱温 4 0℃。结果 :4种药物一次性分离 ,方法回收率均为 97 7~ 98 5 % ,r >0 .999。日内 ,日间RSD均不大于 7%。结论 :本方法简便快速 ,适用于临床血药浓度监测及此类药物中毒的快速定性定量的分析。Objective:To establish a rapid and simple HPLC method for the simultaneous determination of 4 kinds of benzodiazepines including nitrazepam,estazolam,alprazolam and diazepam in serum.Method:The serum samples were extracted with ethyl ether,and the residue was reconstituted in methanol.The mabile phase consisted of methanol acetonitrile water(42∶18∶40 v/v).A discovery C 18 column (250mm×4.6mm,5μm) was used.The detection wavelength was 230nm.Results:4 drugs were separated simultaneously.The coefficients of the linear equation was 0.999.The recovery ratio was 97.74~98.49%,RSD of intra day and inter day are less than 7%.Conclusion:The method was simple and rapid.It was suitable for rapid qualitative and quantitative determination of the toxic drugs as well as the therapeutic monitoring of patients in the serum.
