摘  要:目的 :探讨肝细胞内糖原含量对肝脏缺血再灌注损伤的影响及相关机理。方法 :将 2 1只新西兰大耳白兔分为 3组 :A组 (n =7)禁食 2 4h ;B组 (n=7)标准实验室饮食 :C组 (n=7)在B组基础上再经静脉补充2 5 %葡萄糖液 12 0ml。经上述方法造成 3组兔肝组织糖原含量显著不同 ,然后检测缺血再灌注期间肝组织形态学、肝脏酶学、组织ATP含量、肝细胞内游离Ca2 + 浓度及氧自由基相关指标。结果 :缺血前肝组织中糖原含量越高 ,则在缺血再灌注期间组织结构及肝功受损伤越轻 ,肝组织中ATP水平越高 ,细胞内游离Ca2 + 浓度及氧自由基水平越低。结论 :缺血前增加肘组织糖原含量可显著减轻肝脏缺血再灌注损伤程度 ,提高糖原含量能抑制细胞内游离Ca2 + 超载及氧自由基的产生可是其拮抗缺血再灌注损伤的机理之一。Objective:To investigate the effect of hepatic glycogen content on liver ischemia reperfusion injury and its mechanism.Method:Twenty one New Zealand rabbits were divided into 3 groups:A(n=7)fasting for 24h;B(n=7)standard laboratory diet;C(n=7)standard laboratory diet+25%GS 20ml,in,q4h×6,total 120ml.The changes of histomorphology of liver tissue, liver enzymology,ATP content,intracellular free Ca 2+ concentration([Ca 2+ ] i) and OFR production were measured in this study.Result:In rabbits with higher glycogen content in the diet,the morphological changes of the liver tissue and liver function impairment were mild,the higher was the ATP content,the[Ca 2+ ] i concentration and OFR level would be lower.Conclusion:Increasing hepatic glycogen content could ameliorate ischemia reperfusion injury.from inbibit the overload of free calciumions in the tissue cells and the production of OFR,and with the impression that it might play a part in its mechanism
