摘  要:目的:观察人重组肝细胞生成素(rhHPO)迅速诱导瞬时早期反应基因表达情况.方法:利用PT-PCR方法观察HPO刺激原代培养大鼠肝细胞24h内选择性肝再生相关基因mRNA表达情况.结果:HPO在原代培养大鼠肝细胞体系中可迅速诱导瞬时早期反应基因c-fos,LRF-1d的表达,对PC3和Tec基因也有诱导表达的作用,高峰出现在刺激后的0.5-2h,表达量增加3-5倍.结论:HPO可迅速诱导瞬时早期反应基因表达,可能是HPO促肝细胞增生的分子作用机制,首次报道PC3和Tec基因是一种与肝再生调控密切相关的早期反应基因.AIM:To investgate recombinant human Hepatopoietin (rhHPO) rap idly induces the expression of immediate early gene. METHODS:We investigated the different gene mRNA expression relat ed liver regeneration within 24 h by PT-PCR in primary cultured hepatocytes sys tem. RESULTS:HPO can rapidly induce the expression of immediate early gene (c-fos, LRF-1, PC3 and Tec, etc.) in primarily cultured rat hepatocytes. The levels of c-fos and LRF-1 mRNA ,which are also known to be induced during the early hours of liver regeneration, were increased after treatment of isolat ed hepatocytes with rhHPO. Addition of HPO resulted in rapid and transient induc tion of PC3 and Tec mRNA with a peak 0.5-2 h(3-5fold). CONCLUSION:rhHPO can rapidly induce the expression of im mediate early gene, which might be one of HPO molecular mechanism on stimulatin g hepatocyte proliferation. This is the first report that PC3 gene and Tec gene are immediate early gene related to liver regeneration.
