终末期肾功能衰竭患者ApoE基因多态性的分布及意义

摘　　要：目的研究终末期肾功能衰竭 (ESRF)发生的遗传学倾向 ,寻找与ESRF发生有关的基因。方法对 94例以原发性慢性肾小球肾炎为根底疾病的ESRF患者和 10 8例健康正常人进行载脂蛋白E(ApoE)基因多态性研究。结果ESRF患者ApoE2 / 3基因型 (31 91%VS13 89% ,P <0 0 5 )及ε2等位基因 (2 3 94%VS10 6 5 % ,P <0 0 5 )频率明显高于正常人 ,而ApoE3/ 3基因型 (5 0 %VS70 37% ,P <0 0 5 )及ε3等位基因 (6 9 15 %VS82 41% ,P <0 0 5 )则明显低于正常人。结论ApoE基因多态性与原发性慢性肾小球肾炎的肾功能恶化及肾功能衰竭的发生有一定联系 ,对ApoE基因多态性的分析 ,对预测此类疾病患者预后有一定价值。Objective In order to confirm the tendency of heredity in end stage renal failure(ESRF), and to find out the related genes. Methods ApoE genetic polymorphism was analyzed in 94 patients with ESRF(those patients had chronic glomerulonephritis), and 108 normal control. Results The frequencies of ApoE2/3 genotype and ε2 allele were significantly higher in ESRF patients than those in normal control(ApoE 2/3 : 31.91% vs 13.89%, P <0.05; ε2:23.94% vs 10.65%, P <0.05), while the frequency of ApoE 2/3 genotype and ε3 allele were much lower in ESRF compared with normal control(ApoE 2/3 : 50% vs 70.37%, P <0.05; ε3: 69.15% vs 82.41%, P <0.05). Conclusion ApoE gene polymorphism is associated with the development of chronic renal failure and may be significant for progression of renal disease.
