摘 要:目的 研究脐血浆刺激造血细胞体外生长的作用及脐血浆造血因子的水平。方法 以ELISA法检测 33例脐血浆GM -CSF ,IL - 3和IL - 6水平 ,RIA法检测EPO水平 ,液体培养和甲基纤维素半固体培养方法检测脐血浆对外周造血干细胞 (PBSC)造血刺激活性 ,并分析二者的相关性。结果 ①脐血浆造血因子中位数和全距水平分别为 :GM-CSF为 0ng/L和 0~44ng/L ,IL - 3为 3ng/L和 0~ 35ng/L,IL - 6为 3ng/L和 0~ 2 1ng/L ,EPO为 1 6U/L和 5~ 72U/L ;②脐血浆刺激下PBSC半固体培养造血祖细胞集落数为 (1 3± 1 1 ) / 1× 1 0 5细胞 ,胎牛血清刺激下的集落数为 0 ,二者差异有显著性 (P <0 .0 1 )。脐血浆或胎牛血清刺激下PBSC液体培养有核细胞数分别为 (1 .0 8± 0 .2 0 )× 1 0 6 /ml与 (0 .72±0 .0 5)×1 0 6 /ml,二者差异有显著性 (P <0 .0 5) ;③脐血浆刺激下PBSC体外培养集落数和有核细胞数均与脐血浆IL -3水平成正相关 ,相关系数分别为 0 .660和 0 .397,与IL - 6和EPO则不相关。结论 脐血浆含有多种造血因子 ,在体外具有刺激造血细胞生长的作用。脐血浆刺激造血活性与其所含有的造血因子有关 ,造血活性的高低与IL - 3浓度成正相关。IL - 6和EPO所起的作用则相对较小。Objective To evaluate the correlation between the hematopoietic stimulatory capacity of cord blood plasma (CBP) and levels of hematopoietic growth factors in CBP. Methods GM-CSF, IL-3 and IL-6 levels were measured by enzyme-linked immunosorbent assay (ELISA). EPO level was measured by radioimmunoassay (RIA). Hematopoitic stimulatory capacity of CBP was represented by colonies in methylcellulose culture of peripheral blood stem cells(PBSCs) with CBP and by numbers of nucleated cells in liquid culture of PBSCs with CBP. Correlation was analysed. Results ① Plasma median levels and ranges were 3 ng/L and 0~35 ng/L for IL-3 in CBP, 3 ng/L and 0~21 ng/L for IL-6, 0 ng/L and 0~44 ng/L for GM-CSF,and 16 U/L and 5~72 U/L for EPO. ② The CFU yields of PBSCs stimulated by CBP or FCS were (13±11)colonies/1×10 5 cells vs 0 colonies/1×10 5 cells ( P <0.01), and the numbers of nucleated cells in liquid media supplemented with CBP or FCS were (1.08±0.20)×10 6/ml vs (0.72±0.05)×10 6/ml ( P <0.05). ③ Positive correlation was found between the yields of colonies stimulated by CBP the levels of IL-3 in CBP. Positive correlation was also found between the numbers of nucleated cells stimulated by CBP and the levels of IL-3. No correlation with IL-6 or EPO was found. Conclusion CBP contains several hematopoitic growth factors and hematopoietic stimulatory activities. The hematopoietic stimulatory activities of CBP required synergistical actions of hematopoietic growth factors, especially IL-3.
