摘  要:目的 研究B族链球菌 (GBS)荚膜多糖 破伤风类毒素结合疫苗主动免疫孕鼠产生型特异性IgG抗体对新生小鼠致死性GBS感染的保护作用。方法 以Ⅲ型GBS荚膜多糖 破伤风类毒素结合物免疫孕鼠 ,用ELISA法测定母鼠型特异性IgG抗体的产生情况及新生鼠体内抗体水平 ,然后以同型GBS诱导新生鼠致死性感染 ,观察其死亡率。孕鼠注射生理盐水 ,新生鼠以致死剂量GBS攻击组作为感染对照组 ,孕鼠及新生鼠不给任何处理组作为正常对照组。结果 接受免疫的孕鼠在临产前型特异性抗体水平大幅度升高 ,几何均数由 0 0 39mg/L升至 0 72 8mg/L (P <0 0 0 1) ,其所产新生鼠体内抗体含量显著高于感染对照组 ,二者的几何均数分别为 0 5 38mg/L、0 0 43mg/L (P <0 0 0 1) ;GBS致死性感染后死亡率显著低于感染对照组 ,分别为 37%、97%(P <0 0 0 1)。正常对照组中新生鼠存活率为 10 0 %。结论 GBS荚膜多糖蛋白结合物疫苗主动免疫孕鼠能诱导型特异性抗体大量产生 ,新生小鼠从母体获得的特异性抗体能保护其免受同型GBS致死性感染 ,体液免疫在新生小鼠的抗GBS感染免疫中起重要作用。Objective Group B streptococcus (GBS) is one of the important pathogens which cause neonatal meningitis, neonatal sepsis and neonatal pneumonia. It even can cause neonate death. In order to find out the methods of prevention and cure the diseases caused by GBS, we investigated the protection of type-specific antibody induced by active immunizing the pregnant mice with GBS type Ⅲ polysaccharide tetanus toxoid conjugate vaccine to newborn mice against lethal GBS infection. Methods Twenty SPF CD-1 pregnant mice were divided into three groups randomly. There were 8 pregnant mice in group A and B, 97 and 99 newborn mice in the two groups, respectively. Active immunize the pregnant mice in group A by injecting intra-peritoneally with 10 μg of GBS type Ⅲ polysaccharide-tetanus toxoid conjugate vaccine was carried out. In group B, the injection control group, the pregnant mice were injected with normal saline. While in group C there were 4 pregnant mice and 56 newborn mice not given any procedure served as normal control. There were 32 newborn mice both in group A and B served as samples for detecting the level of the antibody. Sixty-five (97 minus 32) and 67 (99 minus 32) newborn mice in group A and group B, respectively were challenged with lethal dose of the same type of GBS. Bacterial culture from spleen tissue was made in every dead mouse in group A and B. The type-specific antibody of the mother due to the immune response and the level of the same antibody in the newborn mice were measured with ELISA method. Results The amount of type-specific antibody in the pregnant mice in group A increased greatly after immunization. The geometric mean of the level of the antibody was increased from 0.039 mg/L before immunization to 0.728 mg/L after immunization (P<0.001). The levels of this kind of antibody in newborn mice were higher than the infection control group, which was 0.538 mg/L and 0.043 mg/L, respectively (P<0.001).
