摘  要:目的 在原核系统中表达并纯化人白细胞介素 18(hIL 18) ,制备兔抗人白细胞介素 18多克隆抗体。方法 用RT PCR扩增出hIL 18的cDNA ,克隆到原核表达载体pJW2中 ,转化大肠杆菌JM10 1中诱导表达并纯化。用纯化的人白细胞介素 18免疫新西兰兔制备多克隆抗体。结果 克隆得到序列正确的hIL 18cDNA ,与载体连接后转化入大肠杆菌JM10 1中诱导表达并成功纯化 ,并用纯化的人白细胞介素 18成功制备了兔抗人白细胞介素 18多克隆抗体。Objective To express and purify the recombinant human interleukin 18 (rhIL 18) in prokaryotic system and to prepare the rabbit anti human interleukin 18 polyclonal antibody.Methods The cDNA of hIL 18 was amplified by RT PCR and was cloned into the expression vector pJW2 and transformed into the E.coli JM101. The purified recombinant protein was used to immune the rabbit for preparing polyclonal antibody.Results The cDNA of hIL 18 was sequenced and transformed into the E.coli. The recombinant human IL 18 (rhIL 18) was expressed efficiently in inclusion bodies and was successfully purified, the polyclonal antibody was successfully prepared with the purified recombinant human IL 18.Conclusion The rabbit anti human interleukin 18 polyclonal antibody can be used in further studies.
