摘  要:目的:研究抗藻酸盐血清对粘液型铜绿假单胞菌粘附性及其生物被膜渗透性的影响。方法:采用结晶紫染色法测定粘液型铜绿假单胞菌的粘附性,采用HPLC法测定加替沙星在粘液型铜绿假单胞菌生物被膜的渗透浓度,从而观察抗藻酸盐血清对于粘液型铜绿假单胞菌生物被膜渗透性的影响。结果:1:1抗藻酸盐血清在同 8X MIC加替沙星联用的情况下,使细菌对硅胶片粘附性显著降低,硅胶片洗脱液在540nm波长处的光密度值从0.126 ± 0.011降低到0.114±0.010;同时使加替沙星对细菌生物被膜的渗透性增强,4,8和16mg·L-1 的加替沙星通过细菌生物被膜的浓度分别从1.210±0.091,2.911±0.112和5.511±0.213mg·L-1显著提高至1.752±0.122、3.908±0.154和7.898±0.321mg·L-1。结论:抗藻酸盐血清能够降低粘液型铜绿假单胞菌对于生物医学材料的粘附性,增强抗生素对于生物被膜的渗透能力。OBJECTIVE:To assess the the influence of anti-alginate serum on the adherence of mucoid Pseudomonas aeruginosa biofilm and the penetration of biofilm. METHODS:To determine the adherence of mucoid Pseudomonas aeruginosa by crystal violet stained method; the gatifloxacin penetration concentration was determined by HPLC. RESULT: When 1:1 dilution anti-alginate serum combined with 8 x MIC gatifloxacin, the adherence of mucoid Pseudomonas aeruginosa decreased significantly , the optical density of silicon slides at 540nm decreased from 0.130±0.010,0.129+0.O15andO.126±0.O11toO.120±0.010,0.117±0.O15and 0.1140 ±0.010. The penetrated concentrations of gatifloxacin through biofilm were increased from 1.210±0.091,2.911±0.112 and 5.911±0.213 to 1.752±0.122,3.908 ±0.154 and 7.898 ±0.321 CONCLUSION:The anti-alginate serum could reduce the adherence of Pseudomonas aeruginosa and enhance the penetration ability of gatifloxacin.
