摘 要:目的 :探讨烷化溶血磷脂 (ALP)对HL - 60细胞增殖、凋亡及分化的影响。方法 :克隆形成率测定细胞生长 ;细胞形态学、DNA电泳分析、流式细胞术检测细胞凋亡 ;硝基蓝四氮唑 (NBT)还原实验分析细胞分化。结果 :ALP作用后 ,HL - 60细胞的集落形成率明显下降 (P <0 .0 1) ;15mg/L的ALP作用HL - 60细胞 9h后 ,出现特征的凋亡形态学改变及DNA片段化降解 ;1mg/L的ALP持续作用HL - 60细胞 ,可使NBT还原率提高到 84 .2 %± 2 .6%。结论 :ALP可抑制HL - 60细胞增殖 。AIM: To explore the effect of alkyl-lysophospholipids(ALP) on the proliferation, apoptosis and differentiation of HL-60 cells. METHODS: Proliferative potential was measured by colony formation assays. Apoptotic cells were detected by morphology, DNA gel electrophoresis and flow cytometry analysis. Both morphological criteria and NBT dye reduction were utilized to determine the extent of differentiation. RESULTS: After 9 h of incubation with 15 mg/L of ALP, apoptotic cells, identified by condensened and fragmented nuclei, were present. After 6 days of incubation with 1 mg/L of ALP, the NBT reduction rate in HL-60 cells increased to 84.2±2.6%. CONCLUSION: ALP can induce apoptosis and differentiation, and inhibit growth in HL-60 cells.
