Effects of Peuceldanum Dunn on Hemorrheology and Hemodynamics in Pulmonary Circulation of Pulmonary Hypertensive Rats

摘  要:目的 :评价白花前胡对慢性炎症性肺动脉高压的防治效果 ,探讨肺循环血液流变学特征的改变在肺动脉高压形成中的作用。方法 :采用野百合碱 (monocrotoline,MCT)诱导的慢性炎症性肺动脉高压大鼠模型 ;白花前胡 (5 0 ,10 0 ,15 0mg·kg-1/d)采用管饲给药 3周 ;治疗效果的评价采用血流动力学 (肺动脉平均压 )、血液流变学(表观及还原粘度、红细胞聚积指数 )及相关性分析。结果 :白花前胡可分别使MCT诱导的肺动脉高压降低14.5 %、19.2 % (P <0 .0 5 )、18.8% (P <0 .0 5 )。白花前胡 (10 0mg·kg-1/d)能有效降低大鼠MCT组肺循环血液高切 (2 0 0 /s)表观粘度 2 3.9% (P <0 .0 5 )、低切 (1/s)表观粘度 39.6 % (P <0 .0 1)和红细胞聚积指数 15 .7% (P <0 .0 5 )。其降低表观粘度 (2 0 0 /s ,1/s)与其降低肺动脉压间的相关系数分别是 0 .86 8(P <0 .0 5 )、0 .6 35 (P <0 .0 5 )。结论 :白花前胡能通过改善肺微循环 。Objective:Our purpose was to assess the effects of Peucedanum Dunn abstract (Pd) on monocrotaline (MCT) induced chronic “inflammatory” pulmonary hypertension (PH) and the role of the hemorrheology changes in pulmonary circulation in the development of PH. Methods: Rats models of MCT induced PH were established. Pd (50, 100, 150 mg·kg -1 /d) were administered via gavage for 3 weeks. The therapeutic efficacy was evaluated by hemodynamics (pulmonary artery pressure, PAP), hemorrheology (apparent viscosity and RBC agglutination index, RAI), and correlation analysis. Results: Pd (50, 100, 150 mg·kg -1 /d) inhibited the increase of PAP of MCT induced PH by 14.5%, 19.2% (P<0.05) and 18.8% (P<0.05), respectively. The effect of Pd (100 mg·kg -1 /d) on hemorrheology was analysed. Pd (100 mg·kg -1 /d) decreased the rise of the apparent viscosity at high shear rate (200/s) , low shear rate (1/s), and RAI of MCT treated rats by 23.9% (P<0.05) , 39.6% (P<0.01) , 15.7% (P<0.05), respective ly. The correlation index between the decrease of apparent viscosity (200/s, 1/s) and the decrease of PAP was 0.868 (P< 0.05) , 0.635 (P<0.05), respectively. Conclusion:Pd can effectively prevent the development of chronic “inflammatory” PH in rats by ameliorating of pulmonary microcirculation.
