摘 要:目的 :建立人牙龈沟液中的替硝唑浓度的 HPL C测定。方法 :龈沟液样品用乙酸乙酯在 p H7.0条件下提取 ,常温下氮气吹干 ,残渣用流动相溶解 ,进样 2 0 μl,色谱条件 :Zorbax SB- C1 8柱 (2 5 0 mm× 4.6 mm,5 μm) ;流动相为甲醇 -水 -冰醋酸 (2 0∶ 80∶ 0 .1) ;流速为 1.0 m l/m in,检测波长为 310 nm。结果 :本法在 0 .0 2~ 0 .8μg间线性良好 ,相关系数 r=0 .9998。高中低浓度的 3个质控样品日内日间变异系数 RSD都在 1.7%~ 4.1%之间 ,回收率在 90 .5 %~ 92 .4% ,龈沟液中最低检测量为 5 ng。结论 :本方法能简便。OBJECTIVE:To establish an assay method by HPLC for tinidazole in human gingival crevicular fluid. METHOD:The tinidazole was extracted from gingival crevicular fluid with ethyl acetate at pH7.0. The organic layer was evaporated and the residual was reconstituted with mobile phase. The tinidazole was assayed on a C 18 column with a mobile phase consisting of methanol water glacial acetate acid(20:80:0.1) at a flow rate of 1.0 ml/min and detected at 310nm. RESULTS: The linearity was obstained from 0.02~0.8 μg of tinidazole in gingival crevicular fluid with a good correlation (r= 0.9998 ). The intraday and interday precisions were less than 4.1%,and the mean recoveries were 92.4%,90.5% and 91.7% for low,middle and high concentration samples,respectively(0.04,0.2,0.8 μg).CONCLUSION:The improved method was a sensitive,reproducible and simple assay.
