肾衰冲剂缓解5/6肾切除大鼠肾小球硬化的实验研究

摘　　要：目的:研究扶正降浊的肾衰冲剂缓解慢性肾衰大鼠残余肾小球硬化的作用。方法:制作大鼠5/6肾切除肾衰模型,分别给予肾衰冲剂及主要单味药党参、丹参、制大黄灌胃,治疗1个月后检测大鼠血清Ⅳ型胶原(COL-Ⅳ)、层粘蛋白(LN)、纤维连接蛋白(FN)、Ⅲ前胶原(PC-Ⅲ)含量,图象分析残肾组织Ⅳ型胶原、纤维连接蛋白的表达量。结果:肾衰冲剂可以降低血LN、COL-Ⅳ、PC-Ⅲ前胶原,提高血FN含量,减少残肾肾小球细胞外基质FN、Col-Ⅳ过量积聚。而党参、丹参、制大黄亦有各自一定的作用。结论:肾衰冲剂减轻肾小球硬化的机理在于减少残肾肾小球细胞外基质的增生。Objective:To study the effect of SSCJ amerliorating glomerulosclerosis in remnant kidney on 5/6 SNX rats. Methods:5/6 SNX rats were underlying this experiment.They were adminstered by oral SSCJ, Pilose Asiabell Root,Radix Salviae Miltiorrhizae and Rhei Rhizom respectively.After one month,the following parametres were examined:serum FN、LN、COL-Ⅳ、PC-Ⅲ;the expressions of FNCOL-Ⅳ in remnant kidney were detected by image anlysising in the computer.Results:SSCJ can decrease the amount of serum LN、COL-Ⅳ、PC-Ⅲ, increase the amount of serum FN,decrease the excessively accumlation of ECM of the remnant kidney glomeruli. Pilose Asiabell Root,Radix Salviae Miltiorrhizae and Rhei Rhizom respectively has similar effect at some degree. Conclusion:The mechanism of SSCJ ameliorating GS may be involved in decreasing the hyperplasmia of ECM of the remnant kidney glomeruli.
