摘  要:目的 研究离心法制得胶原膜载释地塞米松的优点 ,探索自制此种胶原膜复合物对眼局部作用的机制。方法 将 2 8只哈白兔左眼结膜下注射地塞米松 2 .5mg(0 .5ml) ,右眼通过胶原膜地塞米松复合物给药 ,按用药后0 .5、1、3、5h随机分为 4组 ,用高效液相色谱法测定兔双眼前房水中地塞米松浓度。结果 左眼地塞米松浓度达到峰值 (1 .882 2± 0 .1 2 89) μg/ml需要 1h,至 5h降到 (0 .0 753± 0 .0 1 1 1 ) μg/ml。右眼给药后房水中药浓度逐渐升高 ,3h达峰值 (1 .0 895± 0 .1 392 ) μg/ml,而 5h维持浓度 (0 .7569± 0 .1 2 35) μg/ml。 结论 胶原膜复合物给药作为无刺激性给药途径起到向眼内缓释并较长时间维持地塞米松眼内高浓度 。Objective To clarify corneal collagen shield as a drug vehicle for the delivery of dexamethasone and to find the mechanism of such collagen shield compound in ophthalmic topical therapy.Methods 28 white rabbits were randomly divided into four groups by times after using dexamethasone.The left eyes of the rabbits were treated with subconjunctival injection,while the right eyes were treated with collagen shield.At 0.5 ,1,3,5 hours after the treatment,the dexamethsone concentration in aqueous humor of rabbits were measured by high pressure liquid chromatography assay.Results The drug concentrations in the left eye's aqueous humor reached the highest peak at 1 hour(1.8822±0.1289)μg/ml.At 5 hours the drug concentration decreased to (0.0753±0.0111)μg/ml.The dexamethasone concentrations of right eye's aqueous humor began to rise at 1 hour and reached the highest peak at 3 hours(1.0895±0.1392)μg/ml.The drug concentrations still kept at (0.7569±0.1235)μg/ml until 5 hours.Conclusion The corneal collagen shield compoud is a noirritate vehicle for the delivery of dexamethasone,while can play a role in dissolving gradually and maintaining high concentration to the eye.The study provide the scientific basis for demonstrating that the collagen shield used in eye can prolong the effective time of the drug.
