摘  要:目的 研究放射损伤小鼠骨髓基质细胞对粒系祖细胞生长的影响。方法 采用体外小鼠骨髓基质细胞贴壁培养法和粒系祖细胞 (CFU GM)培养法。结果 伤后 3~ 7d ,有基质细胞贴壁的CFU GM生长明显低于无贴壁组 ;伤后 14d贴壁层形成后 ,放射损伤组的CFU GM生长明显低于正常对照组 ;放射损伤后 3~ 7d ,骨髓基质细胞可抑制正常CFU GM生长 ,用薄层琼脂分隔基质细胞贴壁层与靶细胞 ,其抑制作用减弱。当无外源性集落刺激因子 (CSF)时 ,基质细胞贴壁层对CFU GM生长仍显示一定刺激作用。结论 辐射使骨髓基质细胞功能受到一定损伤 ,但在放射损伤造血修复与重建过程中 ,骨髓基质细胞仍起着“刺激”与“抑制”Objective To investigate the effects of bone marrow stroma cells from irradiated mice on the proliferation of granulocyte progenitor cells. Methods Health mice were exposed to 60 Gy of γ irradiation every day for 3, 7 and 14 d. The bone marrow nuleated cells of them and the control mice were isolated and cultured for 14 d for stroma cells. The normal, irradiated stroma cells were then co cultured with normal or irradiated murine bone marrow nucleated cells to evaluate the CFU-GM growth. The stimulating effects of stroma cells on the growth of granulocyte progenitor cells were determined. Results The supernatant of cultured normal stroma cells prevented CFU GM growth. Exogenous GM CSF addition to the culture system had no influence on granulocyte progenitor cells. CFU GM growth was obviously inhibited in the medium with nuleated cells form the mice receiving 3 to 7 day irradiation. Agar layer in the culture medium inhibited the growth of CFU GM. Conclusion Bone marrow stroma cells play an important role in the reconstruction of hematopoiesis in mice after being exposed to γ irradiation .
