摘  要:目的 探讨人参皂甙 (Ginsenosides ,GS)对睡眠剥夺 (Sleepdeprivation ,SD)大鼠脑干中缝核群五羟色胺 (Serotonin ,5 HT)的影响。方法 用小平台水环境法建立大鼠SD模型。选用Sprague Dawley大鼠 5 6只 ,随机分为未剥夺睡眠组 (Non SD)、SD 12、2 4、36、48、72、96h组 ,每组又设GS用药组和生理盐水对照组。GS用药组用GS连续灌胃 5d ,然后给予不同时间的SD。SD结束后 ,以4%多聚甲醛灌注固定 ,ABC法组化染色 ,图像分析法分析 5 HT含量 ,用Dλ表示。结果 各组大鼠SD后中缝核群各核团的 5 HTDλ值均升高 ,48h以前升高较慢 ,72h后明显升高。未剥夺睡眠组大鼠 ,用药组Dλ 显著高于对照组 (P <0 .0 5 ) ;SD 48h ,两组无显著性差异 (P >0 .0 5 ) ;SD 72、96h ,用药组显著低于对照组 (P <0 .0 5 ) ;SD 12、2 4和 36h ,各核团的Dλ值变化不一 ,其中在中缝隐核 ,用药组显著高于对照组 (P <0 .0 5 ) ;在中缝大核和中缝正中核 ,两组无显著性差异 (P >0 .0 5 ) ;在中缝背核 ,12和 2 4h ,用药组显著高于对照组 (P <0 .0 5 ) ,36h ,两组无显著性差异 (P >0 .0 5 ) ;在中缝苍白核和中缝桥核 ,12h用药组显著高于对照组 (P <0 .0 5 ) ,2 4和 36h ,两组无显著性差异 (P >0 .0 5 )。结论 SD后 ,大鼠中缝核群 5Objective To investigate the effects of ginsenosides(GS) on rats' serotonin of raphe ceruleus during sleep deprivation(SD). Methods SD was induced in male Sprague Dawlay rats by using 'Flower Pot' technique. Fifty six rats were divided into 7 groups randomly: Non SD(not deprived of sleep), 12SD(deprived of sleep for 12 h), 24SD(for 24 h), 36SD(for 36 h), 48SD(for 48 h),72SD(for 72 h),and 96SD(for 96 h) , and each group was divided into two subgroups: experimental and control. Forced feeding of GS to the experimental rats for 5 d before the rats were deprived of sleep. Then the SD rats were fixed with paraformaldehyed perfusion,and stained with ABC staining method. Rats' serotonin of raphe ceruleus was evaluated by using immunocytochemistry and image analysis system. Results Among all groups, the optical density ( D λ) of raphe ceruleus increased following SD, and the D λ increased rapidly after 72 h of SD. Compared with controls, the D λ of the experimentals increased significantly in Non SD groups( P <0.05), while decreased significantly in 72SD and 96SD groups ( P <0.05). There were no obvious difference between the experimentals and the controls in 48SD groups( P >0.05). Different changes on the D λ of raphe ceruleus were observed in 12SD, 24SD and 36SD groups. Compared with controls, the experimentals′ D λ of raphe obscurus nucleus(ROb) increased significantly in 12SD, 24SD and 36SD groups ( P <0.05), and no difference between experimentals and controls in raphe magnus nucleus (RMg) and median raphe nucleus (MnR) ( P >0.05); the experimentals′ D λ of dorsal raphe nucleus(DR) increased significantly in 12SD and 24SD groups ( P <0.05), and no difference in 36SD groups ( P >0.05); the experimentals′ D λ of raphe pallidus nucleus(RPa) and raphe pontis nucleus(RPn) increased significantly in 12SD groups( P <0.05), and no difference observed in 24SD and 36SD groups ( P >0.05). Conclusion Seretonin significantly increases after SD
