摘 要:目的 探讨半胱氨酸白三烯 1受体拮抗剂孟鲁司特对慢性支气管炎大鼠气道粘蛋白高分泌的作用。方法 气管内注入 0 .2mg脂多糖复制模型 ,采用图像分析、ELISA方法检测病理学改变、粘蛋白表达水平、支气管肺泡灌洗液 (BALF)中粘蛋白量。结果 3周后出现慢性支气管炎的病理学特征 ,如杯状细胞、粘膜下腺体增生肥大 ,腺体细胞和气管上皮粘蛋白面积 ( % )、未分泌指数及BALF中粘蛋白量增加 ;孟鲁司特可显著抑制杯状细胞增生 ,降低分泌细胞粘蛋白表达水平、BALF中粘蛋白含量。结论 孟鲁司特可以在杯状细胞增生、粘蛋白合成、分泌等环节 。Objective To investigate the inhibition of montelukast on hypersecretion of airway mucins in rats with chronic bronchitis. Methods Rat model was established by intratracheal instillation of lipopolysaccharide 0.2 mg. The changes of pathology, mucins expression levels, and mucins content in bronchoalveolar lavage fluid BALF were observed with image analysis and ELISA. Results The rats shared many features of chronic bronchitis weeks after instillation, such as goblet cells, submucosal glands hyperplasia and hypertrophy, increases of mucins area (%) of glands and trachea epithelium pre secretion index and mucins overabundance in secretory cells and BALF. Montelukast remarkably inhibited goblet cells hyperplasia, decreased mucins area (%) of glands and trachea epithelium, pre secretion index, and mucins content in BALF. Conclusions Through inhibiting goblet cells hyperplasia, excessive mucin synthesis and secretion, montelukast can inhibit the hypersecretion of airway mucins.
