摘  要:应用焦锑酸盐沉淀技术对水稻花后衰退珠心和胚乳发育初期进行了Ca2 + 的超微细胞化学定位。结果显示在初始衰退的珠心细胞中Ca2 + 主要分布于液泡膜上和核内 ;在衰退中期的珠心细胞中 ,Ca2 + 主要分布在核膜、液泡膜及质膜上 ;在严重衰退的珠心细胞 ,Ca2 + 仅存在于液泡中。珠心降解的Ca2 + 跨过胚囊壁 ,通过质外体向胚乳内运输 ;发育初期的胚乳细胞 ,Ca2 + 主要位于胞间隙 ,线粒体和液泡中也有少量分布。讨论了Ca2 +Ultracytochemical localization of calcium was carried out using a potassium pyroantimonate precipitation method in the degenerating nucellus and early endosperm cells of rice ( Oryza sativa L) after flowering. The results revealed that calcium deposits were localized on the vacuolar membrane and in the nucleus of nucellar cells at the early degenerative stage, and also observed on the plasma membrane including vacuolar membrane and nuclear envelope at the mid degenerative stage. But calcium was only observed on the vacuolar membrane in the extremely degenerated nucellar cells. The calcium was transported to endosperm cells from degenerating nucellar cells through the embryonic sac wall. In the endosperm cells, the calcium deposits were mainly localized in the intercellular space, whereas few deposits could be observed in the mitochondrium and vacuole. Calcium was transported in the endosperm cells via apoplast system. The relationship between calcium and PCD of nucellar cell was discussed.
