摘  要:目的 了解一株自发突变的宋内志贺菌S7株作为活疫苗受体的安全性。方法 应用肠道细菌毒力和侵袭力检测、宋内菌大质粒DNA图谱分析和猴体口服耐受试验对S7株、S7R株进行安全性和稳定性分析。结果 S7株能很好的表达宋内O抗原 ,有免疫原性。毒力、侵袭力试验阴性 ;细菌侵入HeLa细胞的侵袭百分率为 4.3% ,毒株S5 12为 83.6 % ;Westernblot结果表明无 4种侵袭性外膜蛋白 (IpaA、B、C、D) ;质粒稳定性试验表明 ,连续传代 36次 ,Ⅰ相大质粒仍稳定存在。S7含的Ⅰ相大质粒、猴体口服实验等与宋内毒株S5 12有明显的区别。结论 宋内菌S7株作为活疫苗受体是安全的 ,其形态的变化代表抗原的有无 ,标志明显 。Objective To study on the safety of a spontaneous mutant strain of Shigella sonnei S7 used as a living bacterial vaccine vector. Methods Making use of enterobacterial virulence and invasive test, DNA profile analysis and challenge test of rhesus monkey to determine the safety and stability of S7 and S7R. Results S7 can express high level S. sonnei O antigan and have immunogenicity; the invasive rate to HeLa cell was 4.3% and a virulent strain S. sonnei strain S512 was 83.6%. S7 did not express IpaA, B, C, D in Western bloting, and the large plasmid of S7 was stable after 36 passages. The phase Ⅰ large plasmid in S7 and the chalenge test in monkeys were different significantly from those of S512. Conclusion The strain S7 of S. sonnei as a living bacterial vaccine vector has its advantage. It is safe and its colony morphologic change can reflect the vector being with or without antigenicity.
