摘　　要：目的　观察“高效价抗角蛋白自身抗体注射液”(AKAAI)对银屑病和接触性皮炎动物模型的治疗作用 ,为AKAAI进入临床试验提供药效学资料 .方法　制备心得安豚鼠银屑病样皮炎和硫化钠家兔刺激性接触性皮炎的动物模型 ,分别将两种模型的实验动物分为 5组 ,治疗组按 3个剂量(2 .5 ,5 ,1 0 mg· kg- 1)给予实验动物 im AKAAI,同时设立生理盐水组和去除 AK auto Ab的丙种球蛋白组作对照 .定期观察进行大体评分 ,并取皮损标本固定、包埋 ,光镜观察组织学变化 .结果　豚鼠耳廓涂药前后组织学差异显著 . im 1 0d后 ,AKAAI治疗组豚鼠耳廓组织学中银屑病病理现象基本消失 ,且有明显的剂量效应关系 ,而 NS组与 Ig组的组织学改变不明显 .饱和硫化钠溶液所致家兔接触性皮炎的反应强度各组间较一致 ,与对照组相比 ,im给药 5及 7d后 ,AKAAI治疗组大体评分和组织学炎症反应强度分别出现显著性差异 ,并表现出明显的剂量效应关系 .AKAAI治疗组的疗程明显短于对照组 .结论　 AKAAI能明显促进豚鼠耳廓银屑病样皮炎和家兔接触性皮炎的恢复及愈合 ,缩短疗程 ,显示了良好的治疗作用 ,为 AKAAI进行临床试验提供了有力依据 .AIM To explore the effects of anti keratin autoantibody injection (AKAAI) on guinea pig's psoriatic model and rabbit's contact dermatitic model. METHODS After the guinea pig's psoriatic model and rabbit's contact dermatitic model were prepared, animals of each kind model were divided into 5 groups: 2.5, 5, 10 mg·kg -1 AKAAI as therapeutical groups, NS group and immunoglobulin without AK auto Ab (Ig) as control. Marks of inflammatory grade of animal skin were recorded at given time. Biopsy of skin specimen were followed histologically under light microscope. RESULTS There was a significant difference of guinea pig ear skin's histology before and after local application of propranolol. Ten days after treatment with AKAAI psoriatic histological features of guinea pig auricle skin in AKAAI groups almost disappeared in a dose dependent manner, however, that in NS and Ig group showed no significant changes. Inflammatory intensity of contact dermatitis induced by sodium sulfide among 5 groups was well distributed. Compared with control groups, marks of animal and histology in AKAAI groups presented a dose dependent significant difference 5 d and 7 d respectively after treatment with AKAAI, and course of treatment in AKAAI group was shorter than that in control groups'. CONCLUSION AKAAI could promote the recovery and healing of guinea pig auricle psoriatic dermatitis and rabbit contact dermatitis, shorten the course of treatment, which indicates positive therapeutical effect of AKAAI.
