摘 要:目的 :建立一种测定甘草酸注射液中甘草酸含量的高效毛细管电泳法。方法 :10 mmol.L- 1 磷酸缓冲液 (p H 7.0 )中含12 .5 mm ol.L- 1十二烷基硫酸钠 (SDS)为电泳电解质 ,电压 30 k V,检测波长 2 5 4 nm,毛细管有效长度 5 0 cm,管径 5 0 μm。氨茶碱为内标。结果 :甘草酸与氨茶碱的迁移时间分别为 4 .8和 2 .8m in。甘草酸浓度为 6 2 .5~ 10 0 0 mg.L- 1时 ,其浓度与峰面积呈良好线性关系 ,相关系数 (r)为 0 .9997。日内、日间变异系数均小于 5 .0 %。结论 :该方法简单、快速、灵敏 ,重现性良好 。OBJECTIVE: To develop a method for the determination of glycyrrhizin content in glycyrrhizinate injection with high performance capillary electrophoresis (HPCE). METHOD: Phosphate buffer (10 mmol/L,pH=7.0) and SDS ( 12.5 mmol/L) were used as electrolysate. Aminophylline was used as internal standard at a constant voltage of 30kV (current of 40 50μA) and detected at a wavelength of 254nm. The effective length of capillary is 50cm with a diameter of 50μm. The developing time was 9 min. RESULTS: The migration time for glycyrrhizin and aminophilline was 4.8 and 2.8 min, respectively. A good linearity was shown between the concentration and peak area in the concentration range of 62.5~1000mg.L -1 , r=0.9997. The relative standard deviation for intra day and inter day was less than 5.0%. CONCLUSION: It seemed that the HPCE method is faster, more convenient, more sensitive and las better reproduction with much practical usage.
