摘　　要：目的　探讨重组抗体蛋白的复性规律 .方法　用共溶剂氧化还原、表面活性剂介导、变性剂介导及抗原介导复性四种条件对抗人 TNF- α单域抗体、单链抗体 GST融合蛋白进行了分离纯化 .结果　复性产物的可溶率为 1 0 %左右 .结论　几种复性方法均可使目的蛋白复性 。AIM To analyze the impact factors in the refolding of recombinant antibody. METHODS Under several refolding conditions, three recombinant antibodies of mouse anti hTNFα were purified. RESULTS About 10% of active products were obtained. CONCLUSION The several conditions are good enough to refold the three recombinant antibodies, whereas the best is hTNFα affinity chromatography. Concentration of protein before refolding, speed of refolding, pH and ionic intensity of reagent may be important impact factors.
